摘要
目的观察前列腺癌组织中DJ-1蛋白和单酰甘油酯酶(MGLL)的表达情况,并分析两者与前列腺癌临床病理特征及预后的关系。方法收集2019年8月至2021年8月收治的112例经穿刺病理确诊为前列腺癌的患者作为研究对象。采用蛋白印迹法检测前列腺癌组织和癌旁组织中DJ-1和MGLL的表达水平,分析DJ-1、MGLL与前列腺癌临床病理特征的关系。采用受试者工作特征(ROC)曲线评估患者复发/死亡时DJ-1、MGLL的最佳截断值。术后随访至2023年8月,观察DJ-1、MGLL表达与前列腺癌患者术后2年无复发生存率的关系。结果前列腺癌组织中DJ-1和MGLL的相对表达量分别为0.70±0.09和0.22±0.06,在癌旁组织中分别为0.23±0.03和0.84±0.09,差异均有统计学意义(P<0.05)。前列腺癌组织中DJ-1、MGLL表达水平与TNM分期和淋巴结转移有关(P<0.05),与年龄、肿瘤直径和Gleason评分无关(P>0.05)。ROC曲线显示,患者复发/死亡时,DJ-1和MGLL的最佳截断值分别为0.48和0.22,112例前列腺癌患者术后均获随访,DJ-1高表达患者的术后2年无复发生存率为65.6%,低于DJ-1低表达患者的93.8%(P=0.033);MGLL低表达患者的术后2年无复发生存率为65.2%,低于MGLL高表达患者的90.0%(P=0.041)。结论前列腺癌组织中DJ-1高表达、MGLL低表达,DJ-1、MGLL表达水平与TNM分期、淋巴结转移及无复发生存时间有关。
Objective To observe the expression of human protein DJ-1(DJ-1)and monoacylglycerase(MGLL)in prostate cancer tissues,and analyze their relationship with clinicopathological features and prognosis of prostate cancer.Methods A total of 112 patients diagnosed with prostate cancer by clinical puncture from August 2019 to August 2021 were enrolled as the study objects.Western blotting method was used to detect the expression level of DJ-1,MGLL in fresh cancer tissues and adjacent tissues.The relationship between clinicopathological features of prostate cancer and DJ-1,MGLL was analyzed.Receiver operating characteristic(ROC)curve was used to evaluate the cut-off of DJ-1 and MGLL at relapse or death.Postoperative patients were followed-up to August 2023.The correlation between DJ-1 and MGLL expression and the 2-year recurrence-free survival rate of prostate cancer patients was further analyzed.Results The relative expression levels of DJ-1 and MGLL in the adjacent tissues of prostate cancer were 0.23±0.03 and 0.84±0.09,and those in cancer tissues were 0.70±0.09 and 0.22±0.06,with statistically significant difference(P<0.05).The expression levels of DJ-1 and MGLL in prostate cancer tissues were correlated with TNM stage and lymph node metastasis(P<0.05),but not with age,tumor diameter and Gleason score(P>0.05).ROC curve showed that the cut-off of DJ-1 and MGLL was 0.48 and 0.22 respectively at relapse or death.All 112 prostate cancer patients were followed-up after surgery,and the 2-year relapse-free survival rate of prostate cancer patients with high DJ-1 expression was 65.6%,lower than 93.8%of patients with low DJ-1 expression(P=0.033).The 2-year relapse-free survival rate of prostate cancer patients with low MGLL expression was 65.2%,lower than 90.0%of prostate cancer patients with high MGLL expression(P=0.041).Conclusion The expression levels of DJ-1 and MGLL are correlated with TNM stage,lymph node metastasis,and relapse-free survival in prostate cancer patients.
