摘要
目的:对不同储藏包装条件不同规格的牛膝进行化学成分及色度的定量分析,明确牛膝炮制过程中色度与成分的关联性及质量变化规律。方法:使用紫外分光光度计和高效液相色谱法(HPLC)测量不同规格牛膝在不同储藏包装条件下的色度值和指标性成分β-蜕皮甾酮、25R-牛膝甾酮、25S-牛膝甾酮、齐墩果酸及水分的含量,借助灰色关联分析色度与各指标性成分之间的相关性。结果:从饮片含水量综合考虑,优选低温抽真空包装;从甾酮类成分含量考虑,优选低温抽真空包装;从齐墩果酸成分含量考虑,高温充氮气包装较好;从饮片外观和色度值考虑,优选低温环境储藏。灰色关联分析得出饮片的含水量与其色度值关联最为密切,其次是β-蜕皮甾酮。结论:储藏温度因素对牛膝化学成分的影响较大,而含水量与色度的关联最密切。色度值可以为牛膝储藏程度鉴别和成分含量预测提供参考。最佳的牛膝饮片储藏条件是低温抽真空包装。
Objective:To quantitatively analyze the chemical composition and chromaticity of Niuxi with different stroage,packing and specifications,and to clarify the correlation and change rule of the chromaticity and composition of Niuxi during processing.Methods:The colorimaticity value and index component contents of β-ecdysterone,25R-oxanosterone,25S-oxanosterone and oleanolic acid of Niuxi were measured by ultraviolet spectrophotometer and high performance liquid chromatography(HPLC)under different storage and packaging conditions,and the correlation between the chromaticity value and index component was analyzed by gray correlation analysis.Results:From the comprehensive consideration of the water content of the decoction pieces,low-temperature vacuum packaging was preferred;considering the content of steroid components,low-temperature vacuum packing was preferred;considering the content of oleanolic acid components,high-temperature nitrogen package was better;from the appearance and chroma value of the decoction pieces,it was preferred to store in a low-temperature environment.Grey correlation analysis showed that the water content of decoction pieces was most closely related to its chroma value,followed byβ-ecdysterone.Conclusion:The influence of storage temperature on the chemical composition of Niuxi is great,while the correlation between water content and chroma is the closest.The chroma value can provide reference for the identification of storage degree and the prediction of component content of Niuxi.The best storage condition for Niuxi decoction pieces is low-temperature vacuum packaging.
