摘要
优化AB-8大孔树脂纯化凉粉草总黄酮的工艺,并分析其成分,同时考察其对小鼠运动性疲劳的影响。结果显示:最佳工艺条件为采用70 mL质量浓度1.7 mg/mL、pH 4.2的上样液以1.0 mL/min上样后,使用体积分数73.6%乙醇溶液80 mL以1.5 mL/min流速洗脱,产物中总黄酮含量达到68.1%±0.2%。凉粉草总黄酮类成分包含槲皮苷、山奈酚、异槲皮苷及紫云英苷。动物试验表明,凉粉草总黄酮可延长小鼠的力竭游泳时间和爬杆时间,降低小鼠血清中乳酸与尿素氮含量,增强小鼠肝脏组织中超氧化物歧化酶与谷胱甘肽过氧化物酶活力,且灌胃相同剂量的总黄酮纯化物比提取物效果极显著,表明最佳纯化工艺有利于提高凉粉草总黄酮的抗运动疲劳活性。
The purification process of total flavonoids from Mesona chinensis using AB-8 macroporous resin was optimized.The components were analyzed and the effect on exercise fatigue in mice was investigated.The results showed that the optimal process conditions were as follows:after using 70 mL of loading sample solution with a mass concentration of 1.7 mg/mL and pH 4.2 to load sample at 1.0 mL/min,80 mL ethanol solution with volume fraction of 73.6%was used to elute at 1.5 mL/min,the content of total flavonoids in the product reached 68.1%±0.2%.The components of total flavonoids from Mesona chinensis contained quercitrin,aempferol,isoquercitrin and astragalin.The animal results showed that the total flavonoids from Mesona chinensis prolonged swimming exhaustion time and climbing pole time of mice,reduced the contents of lactic acid and urea nitrogen in serum of mice,and enhanced the activity of superoxide dismutase and glutathione peroxidase in the liver tissue of mice.Gavage of the same dose of the total flavonoids purified was more effective than the extracts,which indicated that the optimal process was conducive to improving the anti-exercise fatigue activity of the total flavonoids of Mesona chinensis.
