摘要
目的建立高效液相色谱法(high performance liquid chromatography,HPLC)同时检测水果加工品中常见防腐剂、甜味剂和合成着色剂含量的分析方法。方法称取匀质样品,调节pH,混合溶剂提取后高速离心,氮吹、定容过滤待测。以甲醇、10 mmol/L乙酸铵为主要流动相梯度洗脱,经Agilent ZORBAX Eclipse Plus C18色谱柱分离,二极管阵列检测器多波长检测,外标法定量。结果18种目标物在24 min内均获得较好分离,0.2~50.0μg/mL的质量浓度范围内线性相关系数均在0.9999以上,方法检出限为0.062~2.000 mg/kg,定量限为0.188~6.000 mg/kg。在低、中、高3个水平,样品加标回收率为80.0%~119.8%,相对标准偏差低于5.0%。结论该方法适用基质种类较多,前处理简单易行,灵敏度和重复性较好,能准确测定水果加工品中相关食品添加剂的含量,对大量样品中常见食品添加剂的快速筛选具有实用价值。
Objective To develop a method for the simultaneous determination of common preservatives,sweeteners,and synthetic colorants in processed fruits by high performance liquid chromatography(HPLC).Methods The homogenized sample was weighed,the pH was adjusted,the solvent was mixed,and then centrifuged at high speed.The nitrogen was blown and the constant volume was filtered to be measured.Methanol and 10 mmol/L ammonium acetate were separated using an Agilent ZORBAX Eclipse Plus C18 column and determined via a multi-wavelength diode array detector and the external standard method.Results The 18 kinds of target substances were well separated within 24 minutes.The linear correlation coefficients within the concentration range of 0.2–50.0μg/mL were all above 0.9999.The limits of detection of method were 0.062–2.000 mg/kg,and the limits of quantitative were 0.188–6.000 mg/kg.At 3 levels of low,medium,and high,the recoveries of sample spiking was 80.0%–119.8%,and the relative standard deviations were less than 5.0%.Conclusion The method is suitable for a wide variety of substrates,with simple and easy pre-treatment,good sensitivity and repeatability.It can accurately determine the content of related food additives in fruit processed products,and has practical value for rapid screening of common food additives in a large number of samples.
