CGF联合脱细胞真皮基质修复兔皮肤缺损的实验研究

摘要 目的 探讨脱细胞真皮基质(ADM)联合浓缩生长因子(CGF)对兔全层皮肤缺损的修复效果。方法新西兰兔12只,术前取血制作CGF膜,在兔背部制备2 cm×1 cm的全层皮肤缺损,A组:ADM+CGF、B组:ADM、C组:空白对照。观察愈合情况,Ⅰ/Ⅲ型胶原纤维生长情况以及TGF-β1、VEGF的表达情况。结果A、B组在1周时已实现初级愈合,镜下胶原纤维排列整齐,C组在2周时实现初级愈合,镜下以Ⅰ型胶原纤维为主,胶原纤维排列紊乱;1周时,TGF-β1表达量A组> B组> C组(P<0.05);2周时,A、B组TGF-β1的表达下降,此时C组的TGF-β1呈增长趋势;1、2周时,VEGF表达量A组> B组> C组(P<0.05)。结论 ADM联合CGF修复兔全层皮肤缺损,可加快创口愈合速度、改善创口愈合质量;增加VEGF的释放,稳定调节TGF-β1的释放以利于伤口的愈合。 Objective To investigate the repair effect of concentrate growth factor(CGF)combined with acellular dermal matrix(ADM)on full-thickness skin defect in rabbits.Methods The blood of twelve New Zealand rabbits were collected blood before surgery to make CGF membrane,and 2 cm×1 cm full-layer skin defects were prepared on the back of rabbits.Group A:ADM+CGF,Group B:ADM and Group C:blank control.The healing,the growth ofⅠ/Ⅲtype collagen fibers and the expression of TGF-β1 and VEGF were observed.Results In group A and group B,primary healing was achieved at 1 week,the collagen fibers were arranged neatly under the microscope.In group C,primary healing was achieved at 2 weeks.Under microscope,type I collagen fibers were dominant,and the collagen fibers were disordered.At 1 week,the expression level of TGF-β1 in group A>group B>group C(P<0.05);At 2 weeks,the expression of TGF-β1 in groups A and B decreased,and the expression of TGF-β1 in group C showed an increasing trend.At 1 and 2 weeks,the expression level of VEGF in group A>group B>group C(P<0.05).Conclusions ADM combined with CGF can accelerate the healing speed and improve the healing quality of rabbit skin defect,which can also increase the release of VEGF and stabilize the release of TGF-β1 to facilitate wound healing.
