摘要
目的探索麻杏石甘汤通过调控PD-L1/PD-1及PI3K/AKT和RAS-ERK信号通路平衡肺部炎症与免疫微环境紊乱状态的具体机制,为麻杏石甘汤的推广应用提供更多科学依据。方法将小鼠分为空白组、模型组、PD-L1抗体组、PD-1抗体组、麻杏石甘汤+PD-L1抗体组、麻杏石甘汤+PD-1抗体组、麻杏石甘汤组、头孢地尼组8组,每组10只,使用肺炎链球菌D39滴鼻小鼠造模。末次给药后,HE染色检测肺组织炎症程度,Elisa检测肺泡灌洗液中IL-6、TNF-α水平,Western blo检测PD-1、PD-L1、P-AKT、P-AKT、P-RAF1和P-ERK蛋白表达。结果麻杏石甘汤可减少肺部炎症,同时升高PD-1/L1含量,并且下调P-AKT、P-RAF1和P-ERK含量。结论麻杏石甘汤可通过PD-1/PD-L1、PI3K/AKT和RAS-ERK调节炎症反应。
Objective To explore the effect of Maxing Shigan Decoction on regulating PD-L1/PD-1,PI3K/AKT and RAS-ERK signaling pathways to balances pulmonary inflammation and immune microenvironment disorder,thereby providing more scientific basis for the promotion and application of Maxing Shigan Decoction.Methods The mice were divided into 8 groups,including blank group,model group,PD-L1 antibody group,PD-1 antibody group,Maxing Shigan Decoction+PD-L1 antibody group,Maxing Shigan Decoction+PD-1 antibody group,Maxing Shigan Decoction group,and cefdinir group,with 10 mice in each group.The mice were induced by intranasal administration of Streptococcus pneumoniae D39.After the last administration,HE staining was used to detect the degree of lung tissue inflammation,Elisa was used to detect the levels of IL-6 and TNF-αin bronchoalveolar lavage fluid,and Western blotting was used to detect the expression of PD-1,PD-L1,P-AKT,P-AKT,P-RAF1,and P-ERK proteins.Results Maxing Shigan Decoction can reduce lung inflammation,increase PD-1/L1 levels,and down-regulate P-AKT,P-RAF1 and P-ERK levels.Conclusion Maxing Shigan Decoction can regulate inflammatory response through PD-1/PD-L1,PI3K/AKT and RAS-ERK;Maxing Shigan Decoction has more unclarified mechanisms that need to be explored.
