摘要
目的:采用网络药理学方法分析加味麻黄汤治疗支气管哮喘的作用机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)筛选出加味麻黄汤中各味中药的活性成分及药物靶点,并取其与支气管哮喘靶点的交集。利用Cytoscape软件构建“药物-疾病”靶点相互作用网络图,通过STRING数据库辅助分析靶点蛋白质-蛋白质相互作用,再利用DAVID数据库和R语言软件对交集靶点进行基因本体论(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:共筛选出加味麻黄汤206个有效成分,得到有效成分对应基因靶点239个、支气管哮喘基因靶点9574个,加味麻黄汤与支气管哮喘共同靶点186个。蛋白质-蛋白质相互作用网络由15个节点、66条边构成,度值大的节点包括肿瘤蛋白P53、丝裂原活化蛋白激酶(Mitogen-Activated Protein Kinase,MAPK)1、MAPK14、视网膜母细胞瘤蛋白1、癌基因Rel的RAS同源蛋白A、细胞周期蛋白依赖性激酶抑制剂1A、AKT丝氨酸/苏氨酸蛋白激酶1、E3泛素蛋白连接酶MDM2、FBJ骨肉瘤原癌基因、MAPK3、MYC原癌基因、雌激素受体1、Jun原癌基因、低氧诱导因子-1α、细胞周期蛋白D1,这些为治疗支气管哮喘的潜在核心靶点。GO富集分析得出202个分子功能条目、76个细胞组分条目和2310个生物过程条目。KEGG通路富集分析获得加味麻黄汤治疗支气管哮喘的KEGG通路179条。结论:加味麻黄汤可以通过多成分、多靶点治疗支气管哮喘。
Objective:To analyze the action mechanism of the Mahuang decoction(麻黄汤)in the treatment of asthma by the method of network pharmacology.Methods:The active ingredients and drug targets of different Chinese medicines in the Mahuang decoction were screened by TCMSP,and the intersection with asthma targets were obtained.The drug-disease targets interaction network diagram was constructed by Cytoscape software.The target protein-protein interaction was analyzed by STRING database.The intersection target gene ontology(GO)function analysis and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were preformed by DAVID database and R language software.Results:A total of 206 active components of the Mahuang decoction were screened,with 239 corresponding gene targets of the Mahuang decoction,9574 asthma gene targets,and 186 common targets of the Mahuang decoction and asthma.The protein-protein interaction network was composed of 15 nodes and 66 edges,in which nodes with larger degree values included TP53,MAPK1,MAPK14,RB1,RELA,CDKN1A,AKT1,MDM2,FOS,MAPK3,MYC,ESR1,JUN,HIF1A,CCND1.These nodes were potential core targets for the treatment of bronchial asthma.The GO enrichment analysis obtained 202 molecular function items,76 cellular component items,and 2310 biological process items.The KEGG pathway enrichment analysis showed that there were 179 KEGG pathways in the treatment of bronchial asthma with the Mahuang decoction.Conclusion:The Mahuang decoction can treat bronchial asthma through multi-component and multi-target.
