成纤维细胞活化蛋白在消化道恶性肿瘤发生发展中的作用及机制研究进展

摘要 成纤维细胞活化蛋白(FAP)是一种丝氨酸蛋白酶家族的跨膜糖蛋白,具有二肽酶和明胶酶的活性。其在多种人类肿瘤的基质中表达,参与多种信号通路,通过多种机制促进肿瘤细胞的增殖、迁移和侵袭,与多种肿瘤的不良预后密切相关。但目前的研究并未对FAP在消化道恶性肿瘤中的表达进行系统阐述,其具体作用机制、作为肿瘤生物标志物及治疗靶点的研究仍处于初始阶段,基于此,本文就FAP在消化道恶性肿瘤发生发展中的作用和机制研究进展作一综述,以期为预测消化道恶性肿瘤的预后及选择新的治疗靶点提供思路。 Fibroblast activation protein(FAP),a kind of transmembrane glycoprotein in the serine protease family,has an activity similar to dipeptidase and gelatinase.It is expressed in the stroma of a variety of human tumors and is involved in different signaling pathways.It promotes the proliferation,migration,and invasion of tumor cells through a variety of mechanisms.Therefore,it is closely related to the poor prognosis of many tumors.However,current studies have not systematically elucidated the expression of FAP in gastrointestinal malignancies,and research on its specific mechanism of action and role as a tumor biomarker and therapeutic target are still in the initial stage.In this article,the role and mechanism of FAP in the occurrence and progression of gastrointestinal malignancies are reviewed,in order to provide ideas for predicting the prognosis of gastrointestinal malignancies and selecting new therapeutic targets.
