摘要
建立了高灵敏同时检测人体毛发中四氢大麻酚(THC)及代谢物四氢大麻酸(THC-COOH)的酶联免疫方法(ELISA)。制备了高亲和性单克隆抗体,并简化了毛发样本前处理步骤。结果显示,样本前处理时间缩短至10 min,THC和THC-COOH的检出限分别为0.52 ng/mL和0.08 ng/mL,样本的加标回收率为92.4%~99.62%。592份毛发盲样的ELISA检测结果与液相色谱-串联质谱法(LC-MS/MS)结果一致性良好,为人体毛发中大麻素类物质的免疫学检测提供了有力支撑。
A highly sensitive enzyme-linked immunosorbent assay(ELISA)was established for the simultaneous detection ofΔ9-tetrahydrocannabinol(THC)and its metabolite 11-nor-9-carboxy-Δ9-tetrahydrocannabinol(THC-COOH)in human hair.The monoclonal antibody with high affinity was prepared,and the pretreatment steps of hair samples were simplified.The results showed that the pre-treatment time of samples was shortened to 10 min,the detection limits of THC and THC-COOH were 0.52 ng/mL and 0.08 ng/mL,respectively,and the spiked recoveries of samples were 92.4%-99.62%.The ELISA results of 592 blind hair samples were in good agreement with that of liquid chromatography tandem mass spectrometry(LC-MS/MS),which provided a strong support for the immunological detection of cannabinoids in human hair.
