摘要
探究S100A4蛋白在银屑病发病中的作用机制。在HaCat细胞中用S100A4蛋白的抗体进行改进的RNA结合蛋白免疫沉淀高通量测序(iRIP-seq),获得高通量测序数据。根据基因本体分析即GO(gene ontology)分析,从生物学方面对S100A4蛋白结合的靶RNA所在基因进行注释和分类。对两次重复试验中均获得的S100A4蛋白结合峰相关基因的GO功能分析。研究结果显示,这些基因主要富集在转录调控、细胞内应答反应、翻译起始等相关通路,包括如RNA聚合酶II对转录的负调控、对内质网应激的反应、细胞对热的反应、对有机环状化合物的反应、翻译起始、细胞对未折叠蛋白的反应、伴侣辅助因子依赖的蛋白质重折叠等。研究发现,S100A4蛋白可能通过影响靶RNA的基因表达参与银屑病的发生发展,深入探究S100A4蛋白对靶RNA的调控机制有助于发现新的银屑病治疗靶点。
Explore the mechanism of S100A4 protein in the pathogenesis of psoriasis.Improved RNA binding protein immunoprecipitation high-throughput sequencing(iRIP seq)was performed using antibodies against S100A4 protein in HaCat cells to obtain high-throughput sequencing data.According to Gene Ontology(GO)analysis,annotate and classify the genes containing the target RNA bound to S100A4 protein from a biological perspective.GO functional analysis of S100A4 protein binding peak related genes obtained from two repeated experiments.The results showed that these genes were mainly enriched in transcriptional regulation,intracellular response reactions,translation initiation and other related pathways,including negative regulation of transcription by RNA polymerase II,response to endoplasmic reticulum stress,cell response to heat,response to organic cyclic compounds,translation initiation,cell response to unfolded proteins,protein refolding dependent on chaperone cofactors.This study found that S100A4 protein may participate in the occurrence and development of psoriasis by affecting the gene expression of target RNA.Further exploration of the regulatory mechanism of S100A4 protein on target RNA can help discover new therapeutic targets for psoriasis.
