摘要
目的:优化桔梗多糖的水提取工艺,分析桔梗多糖对桔梗皂苷溶解度的影响。方法:在单因素试验的基础上结合响应面法,以浸泡时间(A)、加液量(B)、提取时间(C)、提取次数(D)为自变量,多糖提取率为响应值,对桔梗多糖水提取工艺进行优化。采用高效液相色谱法以桔梗皂苷D为指标,比较桔梗总多糖和不同醇沉部位多糖对桔梗皂苷溶解度的影响。结果:桔梗多糖的最佳提取工艺为加水20倍量,浸泡24 min后提取67 min,提取2次,在此条件下的桔梗多糖提取率为65.44%,醇沉后桔梗总多糖的产率为35.94%;桔梗总多糖和各醇沉部位多糖都对桔梗皂苷有增溶作用,其中80%醇沉部位多糖增溶效果最好,其次为50%醇沉部位多糖。结论:所建立的桔梗多糖水提取工艺稳定可行,且不同醇沉部位桔梗多糖均对桔梗皂苷具有增溶作用。
Objective:To optimize the aqueous extraction process of platycodon grandifloras polysaccharide and analyze its effect on the solubility of platycodin.Methods:Based on single-factor experiments,the extraction process of platycodon polysaccharide was optimized by response surface methodology using the extraction yield of polysaccharides as the index,and soaking time(A),liquid addition(B),extraction time(C)and extraction times(D)as the factors.Meanwhile,the effect of total polysaccharides and polysaccharides from different alcohol precipitation sites on the solubility of platycodin was investigated by high performance liquid chromatography with platycodin D as an index.Results:The optimal extraction conditions of platycodon polysaccharide were as follows:soaking time was 24 min,liquid addition was 20 times of water,extraction time was 67 min,and extraction times was twice.The extraction yield of polysaccharide was 65.44%under the conditions.The yield of total polysaccharides was 35.94%after alcohol precipitation.All parts of platycodon polysaccharides had solubilization effect on platycodin,and PGP precipitated by 80%ethanol(PGP-D)had the strongest solubility,followed by 50%ethanol precipitation(PGP-A).Conclusions:The extraction process of platycodon polysaccharide optimized by response surface methodology is stable and reliable.The polysaccharides from different alcohol precipitation sites have solubilization effect on platycodin.
