摘要
本研究对提取的鼠疫耶尔森菌(Yersinia pestis)脂多糖(LPS)进行动物实验,观察不同剂量LPS对小鼠一般状态和脏器的影响情况,为研究鼠疫菌LPS对机体的毒性作用机制提供实验数据,对内毒素早期感染的临床诊断提供理论基础。本试验采用常规方法培养鼠疫菌,试剂盒法提取LPS,对提取的LPS进行成分鉴定和SDS-PAGE电泳。小鼠腹腔注射符合实验要求的LPS,观察小鼠的一般状态变化,经处死、解剖、脏器称重后,综合分析LPS对小鼠器官的损伤程度。结果显示,LPS样品纯度为91%,电泳条带分布在10~17 Ku之间。腹腔注射鼠疫菌LPS可致小鼠出现活动减少,蜷缩无力,被毛松乱和昏睡等症状,肝脏和脾脏肿大、充血现象严重。随着LPS注射剂量的增加,试验组小鼠脏器损伤程度也不断加重,但在注射剂量为450μg/只时,其肝脏湿重较注射剂量为350μg/只和250μg/只时减少,其脾脏湿重较LPS注射剂量为350μg/只减少。结果表明,一定剂量的LPS可引起小鼠出现脏器损伤、氧化应激和免疫系统过度活化等现象,且随着LPS注射剂量的增加,损伤程度不断加重。
To investigate the effects of different doses of lipopolysaccharide(LPS)extracted from Yersinia pestis on the general state and organs of mice,so as to provide experimental data for studying the toxic mechanism of LPS on humans and theoretical basis for clinical diagnosis of early endotoxin infection.Yersinia pestis was cultured by conventional methods,and LPS was extracted by kit method.The components of the extracted LPS were identified and SDS-PAGE electrophoresis was performed.The mice were intraperitoneally injected with LPS in line with the experimental requirements,and the phenotypic changes of the mice were monitor.The mice were sacrificed,dissected and organs are weighted and the degree of organ damage induced by LPS was counted.The results of this experiment showed that the purity of LPS samples was 91%,and the electrophoretic bands distributed between 10 and 17 Ku.Intraperitoneal injection of LPS of Yersinia pestis could cause mice to show symptoms such as decreased activity,curled up,weakness,fluff and lethargy,and the liver and spleen enlarged and congested.With the increase of LPS injection dose,the degree of organ damage in the experimental group also aggravated.But with LPS injected does 450μg/pcs,the wet weight of liver was lower than that of 350μg/pcs and 250μg/pcs,and the wet weight of spleen was lower than that of 350μg/pcs.A certain dose of LPS can cause organ damage,oxidative stress and immune system overactivation in experimental animals.With the increase of LPS injection dose,the degree of injury aggravated.
