摘要
目的旨在探讨续断壮骨方对胫骨干骨折大鼠Hedgehog信号通路的影响。方法将60只健康雄性3月龄无特定病原(Sprague dawley,SD)大鼠采用随机数字表法将其分为空白组、模型组、低剂量续断壮骨方组、高剂量续断壮骨方组,每组各15只,除空白组外其余各组均制备成胫骨干骨折模型,术后24 h,对照组及模型组灌服3 ml/d生理盐水;低剂量续断壮骨方组灌服40 mg/(kg·d),高剂量续断壮骨方组灌服80 mg/(kg·d),观察比较骨折术后各组第14、28、42天骨矿物质密度值(Bone mineral density,BMD)及骨痂体积,各组末次给药后观察各组大鼠生物力学性能、血管新生情况,采用实时荧光定量聚合酶链式反应(Reverse transcription-polymerase chain reaction,RT-PCR)检测各组大鼠胫骨切片Ihh mRNA、Smo mRNA、Ptch1 mRNA、GliL mRNA表达及Westem blot法检测各组骨痂组织Ihh、Smo、Ptch、GliL蛋白相对表达量。结果与空白组比较,模型组骨痂体积、BMD指数、胫骨最大载荷、新生血管面积、新生血管数量、Ihh mRNA、Smo mRNA、Ptch1 mRNA、GliL mRNA、Ihh、Smo、Ptch、GliL水平降低,差异有统计学意义(P<0.05);与模型组比较,低、高剂量续断壮骨方组骨痂体积、BMD指数、胫骨最大载荷、新生血管面积、新生血管数量、Ihh mRNA、Smo mRNA、Ptch1 mRNA、GliL mRNA、Ihh、Smo、Ptch、GliL水平升高,差异有统计学意义(P<0.05);与低剂量续断壮骨方组比较,高剂量续断壮骨方组骨痂体积、BMD指数、胫骨最大载荷、新生血管面积、新生血管数量、Ihh mRNA、Smo mRNA、Ptch1 mRNA、GliL mRNA、Ihh、Smo、Ptch、GliL水平明显升高,差异有统计学意义(P<0.05)。结论续断壮骨方能够通过上调胫骨干骨折大鼠Hedgehog信号通路发挥促进骨折愈合的作用。
Objective To investigate the effect of Xuduan Zhuanggu Formula on the Hedgehog signaling pathway in rats with tibial shaft fracture.Methods A total of 60 healthy 3-month-old male Sprague Dawley(SD)rats without specific pathogens were divided into a blank group,a model group,a low-dose Xuduan Zhuanggu Formula group,and a high-dose Xuduan Zhuanggu Formula group with 15 rats in each group by the random number table method.The tibial shaft fracture model was prepared in all groups except for the blank group.The control group and the model group were given 3 mL normal saline daily.The low-dose and high-dose Xuduan Zhuanggu Formula groups were administered at 40 and 80 mg/(kg·d),respectively.The bone mineral density(BMD)and the callus volume of the groups were observed and compared 14,28,and 42 days after fracture operation.The biomechanical properties and angiogenesis of rats in each group were observed after the last administration.Real-time quantitative reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the expression of Ihh mRNA,Smo mRNA,Ptch1 mRNA,and GliL mRNA in tibial sections of rats in each group,and the relative expression of Ihh,Smo,Ptch,and GliL proteins in callus tissues of rats in each group were detected by Western blot.Results Compared with those in the blank control group,the callus volume,BMD index,tibial maximum load,neovascular area,number of new blood vessels,and expression levels of Ihh mRNA,Smo mRNA,Ptch1 mRNA,GliL mRNA,Ihh,Smo,Ptch,and GliL in the model group were decreased,and the differences were statistically significant(P<0.05).Compared with those in the model group,these indicators in the low-dose and high-dose Xuduan Zhuanggu Formula groups were increased,and the differences were statistically significant(P<0.05).In comparison with the low-dose Xuduan Zhuanggu Formula group,the high-dose Xuduan Zhuanggu Formula group showed distinctly increased levels of these indicators with statistically significant differences(P<0.05).Conclusion Xuduan Zhuanggu Formula can promote fracture healing in rats with tibial shaft fracture by up-regulating the Hedgehog signaling pathway.
