免疫抑制通过增加血浆中CD96的含量减少了COVID-19重症率

摘要 目的研究旨在确定免疫抑制剂是否能够减少COVID-19的重症化。方法研究以19种免疫抑制剂的全基因组关联分析(genome-wide association studies,GWAS)数据作为暴露,以COVID-19事件的GWAS数据作为结局进行了两样本孟德尔随机化(Mendelian randomization,MR)分析。随后通过两步法MR寻找免疫抑制剂减少COVID-19重症化的可能中介因素。结果免疫抑制剂的使用与COVID-19患者住院率之间无因果关系(P=0.405;OR=0.974,95%CI:0.916~1.036),与重症率之间存在负向因果关系(P=1.09×10^(-7);OR=0.845,95%CI:0.795~0.899)。中介MR分析结果显示CD96可作为免疫抑制剂减少COVID-19重症率的中介因素,中介效应为9.938%。结论免疫抑制剂可降低COVID-19重症率,并寻找到了可能的作用机制,为探索新治疗方案提供了新的方向,同时为COVID-19合并免疫疾病的患者提供了健康指导。 Objective To determine whether immunosuppressants can reduce the severity of COVID-19.Methods Genome-wide association studies(GWAS)data including 19 immunosuppressants was employed as exposures,and GWAS data of COVID-19 events as outcomes,and a two-sample Mendelian randomization(MR)analysis was conducted.A two-step MR approach was then used to identify potential mediators for the immunosuppressants in reducing the severity of COVID-19.Results The use of immunosuppressants showed no causal association with hospitalization rates among COVID-19 patients(P=0.405;OR=0.974,95%CI:0.916~1.036),but there was a negative causal relationship with severity rates(P=1.09×10^(-7);OR=0.845,95%CI:0.795~0.899).Mediator MR analysis showed that CD96 acted as a mediator by which immunosuppressants reduce the severity of COVID-19,with a mediating effect of 9.938%.Conclusion Immunosuppressants can lower the severity of COVID-19 and identify a potential mechanism of action,providing a new direction for exploring treatment options.It also offers health guidance for patients with COVID-19 combined with immune disorders.
