摘要
应用超高效液相色谱-四极杆-飞行时间串联质谱(UPLC-Q-TOF-MS/MS)技术,结合多元统计分析对不同产地的布渣叶药材进行质量差异分析,为布渣叶的产地判别和质量评价提供依据。采用UPLC-QTOF-MS/MS对不同产地的布渣叶样品进行全成分分析,应用主成分分析(PCA)、正交偏最小二乘法-判别分析(OPLS-DA)及聚类分析(CA)对云南昆明、广西玉林和广东阳江3个产地共18批次布渣叶药材进行多元统计分析,以变量重要性投影(VIP>1)和t检验结果(P<0.05)为筛选标准,确定不同产地布渣叶药材中具有显著差异的化学成分。从不同产地布渣叶药材中共鉴定出53个化学成分,以黄酮类、原花青素类、有机酸和生物碱等类型化合物为主,其中N-反式香豆酰酪胺、N-反式阿魏酰酪胺为首次在布渣叶药材中报道。不同产地布渣叶的化学成分差异明显,共鉴定出17个差异化学成分,其含量在不同产地呈现不同的变化规律,可作为产地鉴别的依据。
Ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)combined with multivariate statistical method was used to analyze the quality differences of Microctis Folium from different producing areas,so as to provide basis for the origin discrimination and quality evaluation of Microctis Folium.UPLC-Q-TOF-MS/MS was used to analyze the total components of Microctis Folium samples from different producing areas.The principal component analysis(PCA),orthogonal partial least squares discriminant analysis(OPLS-DA)and cluster analysis(CA)were used to analyze 18 batches of Microctis Folium from Kunming of Yunnan Province,Yulin of Guangxi Province and Yangjiang of Guangdong Province.According to VIP>1 and t test(P<0.05),the chemical components with significant differences in Microctis Folium from different producing areas were screened.A total of 53 chemical components were identified from Microctis Folium of different habitats,which were mainly flavonoids,procyanidins,organic acids and alkaloids.Among them,N-p-trans-coumaroyltyramine and N-trans-feruloyltyramine were reported in Microctis Folium for the first time.The chemical components of Microctis Folium from different producing areas were significantly different,and a total of 17 differential chemical components were identified.17 differential chemical components showed different variation rules in different producing areas,which can be used as the basis for the identification of producing areas.
