巴马小型猪血虚证模型的建立

摘要 目的采用乙酰苯肼(acetylphenylhydrazine,APH)联合环磷酰胺(cyclophosphamide,CTX)建立巴马小型猪血虚证模型。方法12只巴马小型猪随机分为模型组和空白组,每组6只,常规喂养7 d后,空白组不做特殊处理,模型组采用实验第1天、第4天皮下注射APH 50 mg/kg,第5~8天腹腔注射CTX 20 mg/kg的方法建立巴马小型猪血虚证模型,观察两组巴马小型猪一般体征,测定红细胞计数(red blood cell count,RBC)、平均红细胞血红蛋白(mean corpuscular hemoglobin,MCH)、红细胞比容(hematocrit,HCT)、血红蛋白(hemoglobin,HGB)、白细胞计数(white blood cell count,WBC)、单核细胞数量(number of monocytes,MON)、淋巴细胞数量(lymphocyte number,LYM)、中性粒细胞数量(number of neutrophil,NEU)以评价模型是否建立成功。结果模型组巴马小型猪出现精神萎靡倦怠,鼻唇、四肢等部位淡白失泽,被毛蓬乱、干燥粗糙等一系列血虚症状,与空白组比较,模型组RBC、HCT、HGB、WBC、MON、LYM均降低,差异有统计学意义(P<0.05)。结论第1天、第4天皮下注射APH 50 mg/kg,第5~8天腹腔注射环磷酰胺CTX 20 mg/kg的方法可以成功建立巴马小型猪血虚证模型。 Objective To establish a blood deficiency syndrome model in Bama miniature pig by using acetylphenylhydrazine(APH)in combination with cyclophosphamide(CTX).Methods Twelve Bama miniature pigs were randomly divided into model group and blank group,with 6 pigs in each group.After 7 days of routine feeding,pigs in the model group were injected subcutaneously with APH at a dose of 50 mg/kg on the first and fourth day,and then intraperitoneally injected with CTX at a dose of 20 mg/kg from the fifth to the eighth day to establish the blood deficiency syndrome model in Bama miniature pig.The general signs of the two groups of Bama miniature pig were observed,and the red blood cell count(RBC),mean corpuscular hemoglobin(MCH),hematocrit(HCT),hemoglobin(HGB),white blood cell count(WBC),number of monocytes(MON),lymphocyte number(LYM),and number of neutrophil(NEU)were measured to evaluate whether the model had been successfully established.Results The model group of Bama miniature pig exhibited signs of blood deficiency syndrome,such as lethargy,pale mucosa and limbs,and rough and dry hair,compared to the blank group.Additionally,the model group showed a decrease in RBC,HCT,HGB,WBC,MON,and LYM,with statistically significant differences(P<0.05).Conclusion The method of subcutaneous injection of APH at a dose of 50 mg/kg on the first and fourth day,followed by intraperitoneal injection of CTX at a dose of 20 mg/kg from the fifth to the eighth day,can successfully establish a blood deficiency syndrome model in Bama miniature pig.
