羟基咔咯及其金属配合物的合成与抗菌性能研究

摘要 为了寻找结构新颖、杀菌活性更好的光敏剂,以10-(4-羟基苯基)-5,15-二(五氟苯基)咔咯(P-0H)为先导化合物,进而合成了四种金属配合物10-(4-羟基苯基)-5,15-二(五氟苯基)咔咯镓(P-0H-Ga)、10-(4-羟基苯基)-5,15-二(五氟苯基)咔咯铜(P-0H-Cu)、10-(4-羟基苯基)-5,15-二(五氟苯基)咔咯铁(P-OH-Fe)和10-(4-羟基苯基)-5,15-二(五氟苯基)咔咯钻(P-OH-Co)。通过紫外-可见吸收光谱、荧光光谱等表征了P-OH、POH-Ga、P-OH-Cu、P-OH-Fe和P-OH-Co五种化合物的结构,并进一步研究其光动力抗菌性能。结果表明:在光照条件下,P-OH与P-OH-Ga在有光照射下的抗菌活性最强,咔咯中心引入镓离子能够增强其荧光发射强度及在可见光区的吸收;而引入铜、铁、镓离子后发生荧光灭,并且光动力抗菌活性有所减弱。对比之下,发现P-OH-Ga是一种更为优良的光动力抗菌试剂。 In order to search for photosensitizers with novel structure and better bactericidal activity,10-(4-hydroxyphenyl)-5,15-bis(pentafluorophenyl)corrole(P-OH)as the lead compound,four metal complexes had been synthesized successively,including 10-(4-hydroxyphenyl)-5,15-bis(pentafluorophenyl)corrole gallium(P-OH-Ga),10-(4-hydroxyphenyl)-5,15-bis(pentafluorophenyl)corrole copper(P-0H-Cu),10-(4-hydroxyphenyl)-5,15-bis(pentafluorophenyl)corrole iron(P-OH-Fe),and 10-(4-hydroxyphenyl)-5,15-bis(pentafluorophenyl)corrole cobalt(P-OH-Co).The structures of five compounds,P-OH,P-OH-Ga,P-OH-Cu,P-OHFe,and P-OH-Co,were characterized by ultraviolet-visible absorption spectroscopy,fluorescence spectroscopy,etc.,and their photodynamic antibacterial properties were further studied.The results showed that under light conditions,P-OH and P-OH-Ga exhibited the strongest antibacterial activity.The introduction of gallium ions into the corrole center enhanced their fluorescence emission intensity and absorption in the visible light region,while the introduction of copper,iron,and cobalt ions quenched the fluorescence,leading to a decrease in photodynamic antibacterial activity.This study indicated that P-OH-Ga was a more excellent photodynamic antibacterial reagent.
