基于近红外光谱法对不同黄芩中有效成分含量及质量评价

Evaluation of Content and Quality of Effective Ingredients in Different Huangqin(Scutellaria baicalensis)Based on Near-infrared Spectroscopy

摘要 目的探究近红外光谱法对不同黄芩中有效成分含量及质量评价。方法收集不同产地58个样品黄芩进行黄芩苷、黄芩素和汉黄芩素的检测,有效成分通过高效液相色谱法含量测定法进行验证。采用近红外光仪对样品近红外光谱进行采集,样品经内部交叉验证建立预测模型,选择其中6个样品进行外部验证,来预测黄芩药材中黄芩苷、黄芩素和汉黄芩素的含量。结果高效液相色谱方法线性好、精密度好、回收率高,可用于黄芩中黄芩苷、黄芩素、汉黄芩素3种活性成分的含量分析。建立的最优校正模型校正模型内部数据如下:黄芩苷R^(2)=0.994,RMSEC=0.438,RMSECV=2.257;黄芩素R^(2)=0.992,RMSEC=0.452,RMSECV=2.234;汉黄芩素R^(2)=0.995,RMSEC=0.447,RMSECV=2.310。提示建立的黄芩中3种有效活性成分定量校正模型性能较高。58个样品经内部交叉验证建立预测模型。黄芩苷、黄芩素及汉黄芩素预测值的平均回收率为99.40%、99.85%、100.40%。结论近红外光谱法用于测定不同黄芩中有效成分(黄芩苷、黄芩素和汉黄芩素)具有检测迅速方便、费用低、污染小、结果准确等优势。 Objective To explore the content and quality evaluation of effective ingredients in different Huangqin(Scutellaria baicalensis)by near-infrared spectroscopy.Methods Fifty-eight samples of Huangqin(Scutellaria baicalensis)from different habitats were collected for the detection of baicalin,baicalein and wogonin,and the effective components were verified by HPLC assay.The near-infrared spectrum of the samples was collected with a near-infrared spectrometer,and the samples were cross verified internally to establish a prediction model.Six samples were selected for external verification to predict the contents of baicalin,baicalein and wogonin in Huangqin(Scutellaria baicalensis).Results The HPLC method had good linearity,precision and high recovery.It can be used for the content analysis of baicalin,baicalein and wogonin in Huangqin(Scutellaria baicalensis).The internal data of the calibration model of the established optimal calibration model was as follows:baicalin R^(2)=0.994,RMSEC=0.438,RMSECV=2.257;baicalin R^(2)=0.992,RMSEC=0.452,RMSECV=2.234;wogonin R^(2)=0.995,RMSEC=0.447,RMSECV=2.310.The established quantitative calibration model for the three effective active ingredients in Huangqin(Scutellaria baicalensis)had high performance.A prediction model was established for 58 samples through internal cross validation.The average recoveries of predicted values of baicalin,baicalein and wogonin were 99.40%,99.85%and 100.40%,respectively.Conclusion Near infrared spectroscopy has the advantages of rapid and convenient detection,low cost,less pollution and accurate results for the determination of the effective components(baicalin,baicalein and wogonin)in different Huangqin(Scutellaria baicalensis).
