摘要
目的探究香叶木素的止痒作用,为将来开发香叶木素止痒药物及临床应用提供科学依据和理论基础。方法采取动物行为学的方法,选用ICR小鼠作为研究对象,建立化合物48/80急性瘙痒模型,观察香叶木素对小鼠急性瘙痒的止痒作用。结果香叶木素具有止痒作用且能剂量依赖性抑制化合物48/80引起的急性瘙痒,但是不能抑制氯喹引起的非组胺依赖性瘙痒,香叶木素联合TRPV1阻断剂辣椒平对瘙痒的抑制作用与单独给予辣椒平的作用没有差异。TRPA1阻断剂HC-030031可以降低香叶木素对瘙痒的抑制作用。结论香叶木素通过抑制TRPV1/激活TRPA1通道抑制组胺依赖瘙痒。
Objective To explore the antipruritic effect of diosmetin and provide scientific basis and theoretical basis for the development of effective antipruritic drugs and clinical application of diosmetin in the future.Methods ICR mice were selected as the research objects by animal behavior method,and compound 48/80 was given to establish the acute pruritus model,and the antipruritus effect of diosmetin on acute pruritus in mice was observed.Results Diosmetin had antipruritic effect and inhibited acute pruritus induced by compound 48/80 in a dose-dependent manner.Diosmetin could not inhibit chloroquine induced non-histamine dependent pruritus.The anti-pruritus effect of diosmetin combined with TRPV1 blocker capsazepine was not different from that of capsazepine alone.TRPA1 blocker HC-030031 can reduce the anti-pruritus effect of diosmetin.Conclusion Diosmetin inhibits histamine dependent pruritus by inhibiting TRPV1/activating TRPA1 channel.
