组蛋白分子伴侣ASF1在胃癌中的诊断和预后价值

The Diagnostic and Prognostic Value of Histone Chaperone ASF1 in Gastric Cancer

目的:探讨组蛋白分子伴侣ASF1在胃癌中的诊断和预后价值。方法:应用GEPIA数据库和Kaplan-Meier数据库评估ASF1的两个亚型ASF1A和ASF1B在胃癌中的表达情况以及在胃癌中的预后价值。选取本院于2021年6月—2022年6月收治的38例胃癌患者为研究对象,验证信息分析学结果。通过收集胃癌组织和癌旁组织,并采用qRT-PCR、免疫组化、Western Blot分析ASF1A和ASF1B的表达量。对患者进行平均18个月的随访,研究ASF1A和ASF1B与患者预后的关系。结果:信息分析学和入组患者的样本分析结果一致表明,ASF1B的表达量在胃癌组织中显著升高。ASF1B与生存时间存在负相关,即其高表达预示着不良预后。此外,ASF1B的表达与肿瘤大小、淋巴结转移和TNM分期显著相关(P<0.05)。然而,ASF1A在胃癌组织和正常胃组织中的表达未发现明显差异。结论:ASF1B在胃癌组织中高表达,并且与胃癌患者的生存时间和临床病理特征密切相关,表明ASF1B在胃癌的诊断和预后预测方面具有重要价值。

Objective:To explore the diagnostic and prognostic value of the histone chaperone ASF1 in gastric cancer.Methods:The expression levels and prognostic significance of ASF1 subtypes including ASF1A and ASF1B in gastric cancer were assessed using the GEPIA and Kaplan-Meier databases.A cohort of 38 gastric cancer patients treated at our hospital from June 2021 to June 2022 was selected to validate the bioinformatics results.Gastric cancer tissues and adjacent normal tissues were collected,and the expression levels of ASF1A and ASF1B were analyzed using qRT-PCR,immunohistochemistry,and Western Blot.The patients were followed up for an average period of 18 months to study the relationship between ASF1A and ASF1B expression and patient prognosis.Results:Both bioinformatics analysis and patient sample analysis consistently demonstrated a significant upregulation of ASF1B in gastric cancer tissues.Higher expression levels of ASF1B were inversely correlated with survival time,indicating a poor prognosis.Additionally,ASF1B expression was significantly associated with tumor size,lymph node metastasis,and TNM staging(P<0.05).However,no significant differences in ASF1A expression were observed between gastric cancer and normal gastric tissues.Conclusion:The overexpression of ASF1B in gastric cancer tissues,along with its close association with patient survival time and clinical pathological features,suggests that ASF1B holds significant value for the diagnosis and prognosis prediction in gastric cancer.
