摘要
为进一步优化红叶南天竹Nandina domestica′hongye′增殖培养条件,提高组培苗芽的质量,研究以红叶南天竹组培苗为试验材料,研究蔗糖浓度、光照强度、pH值、活性炭用量等因素对红叶南天竹增殖效果的影响,采用Box-Behnken响应面分析法对其增殖培养参数进行优化。结果表明,影响红叶南天竹增殖效果的因素主次顺序为光照强度>活性炭用量>蔗糖浓度>pH值,各因素对红叶南天竹增殖效果存在极显著影响(P<0.01),并利用Design Expert 13软件对试验结果进行回归分析,建立了二项多次回归方程数学模型,得出红叶南天竹最优的增殖培养参数为:蔗糖质量浓度为32.44 g·L^(-1)、光照强度为1309.84μmol·m^(-2)·s^(-1)、pH值5.71、活性炭用量0.35 g·L^(-1),增殖系数预测值可达4.82。经验证,该模型相对误差仅为1.66%,证明具有高拟合度和高可靠性。优化后的增殖系数可达4.74,缩短了继代培养周期,提高了增殖芽苗的质量,完善了其种苗快繁体系。
To further optimize the proliferation culture conditions of Nandina domestica′hongye′and improve the quality of tissue culture seedlings,the tissue culture seedlings of N.domestica′hongye′were used as experimental materials.The effects of sucrose concentration,light intensity,pH value,and activated carbon dosage on the proliferation of the tissue culture seedlings of N.domestica′hongye′were investigated.Box-Behnken response surface analysis was used to optimize the culture parameters.The results showed that the primary and secondary order of factors affecting the proliferation effect of N.domestica′hongye′was light intensity>activated carbon dosage concentration>sucrose concentration>pH value,and each factor had a very significant effect on the proliferation effect of N.domestica′hongye′(P<0.01).The results of the experiment were ana55 lyzed by regression analysis using Design Expert 13 software,and a mathematical model of binomial multiple regression equation was established.The optimal breedingparameters were as follows:sucrose concentration was 32.44 g·L^(-1),light intensity was 1309.84μmol·m^(-2)·s^(-1),pH value was 5.71,activated carbon dosage was 0.35 g·L^(-1),and the predicted proliferation coefficient was up to 4.82.It is proved that the relative error of the model is only 1.66%,which proves that the model has a high fitting degree and high reliability.After optimization,the breeding coefficient reached 4.74,which shortened the subculture cycle,improved the quality of proliferating bud seedlings,and enhanced the rapid propagation system of seedlings.
