摘要
目的:比较不同干燥方法对人工繁育冬虫夏草的总蛋白、总多糖和5种核苷类成分的影响,为筛选人工繁育冬虫夏草的最佳干燥方法提供理论依据。方法:人工繁育冬虫夏草后,分别进行全冻干、半冻干和自然干燥处理,采用BCA法、苯酚硫酸法以及HPLC法分别测定虫草的总蛋白、总多糖和5种核苷类成分的含量。结果:三种干燥方法处理的人工繁育冬虫夏草的总蛋白含量没有显著性差异(P>0.05);自然干燥处理的人工冬虫夏草总多糖含量显著高于全冻干和半冻干(P<0.05);全冻干和半冻干处理的人工冬虫夏草中并未检测出肌苷和胸苷,自然干燥处理的人工冬虫夏草检测到5种核苷类成分。结论:自然干燥处理可较好保留人工冬虫夏草总多糖和核苷类成分,值得作为人工繁育冬虫夏草的适宜干燥方法推广。
Objective:To provide a theoretical basis for selecting the optimal drying method for artificial Cordyceps,which is based on comparing the effects of different drying methods on the total protein,total polysaccharides,and five nucleoside components of artificial Cordyceps.Methods:After artificial breeding Cordyceps was subjected to full freeze-drying,semi freeze-drying,and natural drying respectively.The total protein content was determined using the BCA method.The total polysaccharide content was determined using the phenol sulfuric acid method.And the content of five nucleoside components was determined using HPLC method.Results:There was no significant difference in total protein content among the three drying methods(P>0.05).The total polysaccharide content of artificial Cordyceps processed with natural drying was significantly higher than that with freeze-dried and semi-freeze-dried(P<0.05).Inosine and thymidine were not detected in the fully freeze-dried and semi-freeze-dried artificial Cordyceps,while five nucleoside components were all detected in the naturally dried artificial Cordyceps.Conclusion:Among them,the naturally dried Cordyceps has the highest total polysaccharide content,with a higher content of five nucleoside components,which is worth popularizing as a suitable drying method for artificial breeding Cordyceps.
