摘要
本研究旨在通过比对伊犁马与哈萨克马背最长肌的差异表达基因,筛选与马脂质代谢相关的候选功能基因。以伊犁马和哈萨克马为研究对象,采集所选马匹背最长肌样本,对所采样本进行脂质代谢和转录组检测,筛选脂质代谢差异代谢物及差异表达基因,并对筛选出的差异表达基因进行富集分析。结果显示,与哈萨克马相比,伊犁马背最长肌中共筛选出21种显著下调的脂质差异代谢物;两组肌肉样本中共筛选出718个差异表达基因,与哈萨克马相比,伊犁马中有300个基因表达显著上调,418个基因表达显著下调;差异表达基因显著富集在78个GO条目中,其中与脂质代谢相关的主要有蛋白质转运、有机物运输、细胞质、钙离子结合和ATP酶活性等;KEGG显著富集在41条通路中,其中与脂质代谢相关的通路主要有脂质与动脉粥样硬化、糖尿病性心肌病、丙酸代谢和EGFR酪氨酸激酶抑制剂耐药性等。本研究初步确定了APOA1、MPC1、ACOX1、EGF、VLDLR、GAS6和ABCA1等7个基因参与了马脂质代谢,为马脂质代谢的分子机制研究提供了数据参考。
This study aims to identify functional genes related to lipid metabolism by comparing differentially expressed genes between the longissimus dorsi of Yili horses and Kazakh horses.Samples were collected from the two breeds and subjected to lipid metabolism and transcriptome analysis.Differentially metabolized lipids and expressed genes were identified,followed by GO(Gene ontology)and KEGG(Kyoto encyclopedia of genes and genomes)analysis of the selected genes.The results showed that Yili horses had 21 significantly down-regulated lipid metabolites compared to Kazakh horses.718 differentially expressed genes were screened from the muscle samples,with 300 upregulated and 418 down-regulated in Yili horses compared to Kazakh horses.The differentially expressed genes were significantly enriched in 78 GO terms,mainly related to protein transport,organic substance transport,cytoplasm,calcium ion binding and ATPase activity.The KEGG analysis revealed that the main lipid-related pathways were lipid and atherosclerosis,diabetic cardiomyopathy,propanoate metabolism and EGFR tyrosine kinase inhibitor resistance.Seven genes,including APOA1,MPC1,ACOX1,EGF,VLDLR,GAS6 and ABCA1,were preliminarily confirmed to be involved in horse lipid metabolism,providing data reference for further molecular mechanism research.
