摘要
本研究旨在评价厚朴酚(MAG)在体外和体内对猪流行性腹泻病毒(PEDV)的抗病毒活性。在体外试验中,以40.0µM的MAG处理感染PEDV后的IPEC-J2细胞系,利用实时定量PCR技术分析MAG对PEDV病毒基因及宿主炎症因子基因表达的影响。结果显示,与PEDV组相比,MAG处理显著降低了病毒相关蛋白(S、M、N)基因的表达水平,且同时抑制了多种炎症因子(IL-6、IL-8、TNF-α、IFN-β、Mx1、ISG15)基因表达的上调,表明MAG不仅可直接抑制病毒,还可能通过调节宿主的免疫反应,减轻病毒感染造成的病理影响。在PED治疗试验中监测了仔猪的血清生化指标及相关细胞因子的表达水平,结果显示,相比PEDV组,MAG治疗组仔猪的血清生化指标均恢复至正常范围,同时结肠组织中S、M、N病毒基因及IL-8、IL-6、TNF-α、IFN-β和ISG15的mRNA表达水平均显著降低(P<0.05)。进一步验证了MAG在体内对PEDV感染的抑制作用,提示MAG可能通过增强宿主免疫系统的调节功能来对抗病毒。综上所述,MAG的预处理可在一定程度上对抗PEDV感染,揭示了MAG作为潜在抗病毒干预策略的可能性,并为控制PEDV感染提供了新思路。
This study aims to evaluate the antiviral activity of magnolol(MAG)against porcine epidemic diarrhea virus(PEDV)infection both in vitro and in vivo.In the in vitro experiments,IPEC-J2 cell lines infected with PEDV were treated with 40.0µM MAG,and real-time quantitative PCR was used to analyze the impact of MAG on the expression of PEDV viral genes and host inflammatory factor genes.The results showed that compared to the PEDV group,MAG treatment significantly reduced the expression of viral-related genes(S,M,N)and simultaneously inhibited the upregulation of various inflammatory factors(IL-6,IL-8,TNF-α,IFN-β,Mx1,ISG15).These findings indicated that MAG could not only directly inhibit the virus,but also alleviate the pathological effects of viral infection by modulating the host immune response.Furthermore,in the PED treatment trial with piglets,serum biochemical indices and related cytokine expression levels were monitored.The results showed that compared to the PEDV group,the biochemical indices of the piglets in the MAG treatment group returned to normal ranges,while the mRNA expression levels of S,M,N viral genes and IL-8,IL-6,TNF-α,IFN-β,and ISG15 in colon tissues were significantly decreased(P<0.05).This further confirmed the inhibitory effect of MAG on PEDV infection in vivo,suggesting that MAG might combat the virus by enhancing the regulatory capacity of the host immune system.In conclusion,pretreatment with MAG showed promising efficacy against PEDV infection.These findings suggested MAG as a potential antiviral intervention strategy and offered new insights into controlling PEDV outbreaks.
