HPLC法同时测定喘咳安丸中柚皮苷等3种成分的含量

Establishment of HPLC method for the determination of naringin,neohesperidin and baicalin in Chuankean pills

建立HPLC方法同时测定喘咳安丸中柚皮苷、新橙皮苷、黄芩苷的含量。采用Hypersil ODS2 C18色谱柱(250.0 mm×4.6 mm,5µm);流动相为乙腈(A)-0.1%磷酸溶液(B),梯度洗脱;流速为1.0 mL/min;柱温为35℃;检测波长为283 nm,进样量为10µL。结果显示,柚皮苷、新橙皮苷、黄芩苷分别在0.0430~0.8602µg/mL(r=0.9999)、0.0501~1.0016µg/mL(r=0.9999)、0.0469~0.9386µg/mL(r=0.9999)范围内与峰面积呈良好的线性关系;精密度试验(n=6)、重复性试验(n=6)、稳定性试验(48 h)RSD均小于2.0%,平均加样回收率(n=9)分别为99.54%、99.95%、100.01%,RSD分别为0.29%、0.36%、0.26%。该方法操作简便,结果准确,可为喘咳安丸的质量控制提供科学依据。

The objective was to establish a HPLC method for the determination of naringin,neohesperidin and baicalin in Chuankean pills.The Hypersil ODS2 C18 column(250.0 mm×4.6 mm,5µm)was used,the mobile phase was acetonitrile-0.1%phosphoric acid with gradient elution,the flow rate was 1.0 mL/min,the column temperature was 35℃,the detection wavelength was 283 nm and the sample size was 10µL.The results showed that naringin,neohesperidin and baicalin presented a good linear relationship with peak area in the range of 0.0430-0.8602µg/mL(r=0.9999),0.0501-1.0016µg/mL(r=0.9999)and 0.0469-0.9386µg/mL(r=0.9999),respectively.The RSD of precision(n=6),repeatability(n=6)and stability(48 h)were all less than 2.0%.The average recovery of naringin,neohesperidin and baicalin(n=9)was 99.54%,99.95%and 100.01%with RSD of 0.29%,0.36%and 0.26%,respectively.The method is simple and accurate,and can provide scientific basis for the quality control of Chuankean pills.
