自拟益气养阴方对气阴两虚证大鼠影响

Effects of a Self-proposed Yiqi Yangyin Decoction(自拟益气养阴方)on Rats with Qi and Yin Deficiency Syndrome

目的研究自拟益气养阴方(简称益气养阴方)对气阴两虚证大鼠的影响。方法将48只SD大鼠随机分为对照组,模型组,阳性药组,益气养阴方高、中、低剂量组。采用限食、灌胃热性药、负重游泳的方法制备气阴两虚证大鼠模型,造模时间为2周。造模成功后,给予阳性药(生脉饮)或益气养阴方水煎液治疗,时间为2周。给药期间观察大鼠一般行为学;采用HE染色观察胃、肝、脾组织结构变化;检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、葡萄糖(GLU)、甘油三酯(TG)水平变化;ELISA法检测血清免疫球蛋白A(IgA)、免疫球蛋白G(IgG)、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)水平。结果与对照组相比,模型组大鼠的体质量、尾根部直径和抓力均明显降低(P<0.01);模型组大鼠胃黏膜上皮细胞损伤,胃黏膜固有层腺体排列紊乱且数量减少,部分肝细胞变性、坏死,脾脏红髓面积扩大,白髓面积缩小,血清中AST、ALT、cAMP/cGMP显著升高(P<0.05或P<0.01),GLU、lgA、lgG显著降低(P<0.01)。与模型组相比,益气养阴方可以改善大鼠胃、肝、脾组织结构;阳性药和益气养阴方高、中剂量可明显改善大鼠的一般行为学指标、肝功能、糖脂代谢、免疫功能和环核苷酸系统(P<0.05或P<0.01)。与阳性药组相比,益气养阴方中剂量组大鼠血清AST水平显著降低(P<0.05),益气养阴方高剂量组大鼠血清cGMP含量显著降低(P<0.01)。结论自拟益气养阴方能显著改善气阴两虚证大鼠的症状,机制可能与调节免疫功能、改善环核苷酸系统有关。

Objective To study the effects of Self-proposed Yiqi Yangyin Decoction(自拟益气养阴方)on rats with Qi and Yin deficiency syndrome.Methods Forty-eight SD rats were randomly divided into the control group,the model group,the positive group,and the high,medium and low dose groups of Yiqi Yangyin Decoction.The rat model of Qi and Yin deficiency syndrome was prepared by restricting food,gavage of hot medicines,and weight-bearing swimming,and the rat model needed 2 weeks.After 2 weeks,the positive drug[Shengmai Drink(生脉饮)]or the aqueous decoction of the Yiqi Yangyin formula was given for 2 weeks.The general behavioural indicators of rats were observed during administration;the structural changes of stomach,liver and spleen tissues of rats were observed by HE staining;levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),glucose(GLU)and triglyceride(TG)in serum,were measured;levels of immunoglobulin A(IgA),immunoglobulin G(IgG),cyclic adenosine monophosphate(cAMP),and cyclic guanosine monophosphate(cGMP)in serum were measured by ELISA.Results Compared with the control group,the body weight,tail root diameter and grip strength of rats in the model group were significantly reduced(P<0.01);the epithelial cells of the gastric mucosa of rats in the model group were damaged,and the arrangement of glands in the lamina propria of the gastric mucosa was disorganised and the number of glands was reduced;some hepatocytes were degenerated and necrotic;the area of the red pulp of the spleen was enlarged,and the area of the white pulp was reduced;the levels of AST,ALT,and the ratio of cAMP/cGMP were significantly elevated(P<0.05 or P<0.01);the levels of GLU,IgA,IgG were significantly decreased(P<0.01).Compared with the model group,Yiqi Yangyin Decoction could improve the histological structure of the stomach,liver and spleen of rats;the positive drug and the high and middle doses of Yiqi Yangyin Decoction could significantly improve the general behavioural indicators,liver function,glucose and lipid metabolism,immune function and cyclic nucleotide system of the rats(P<0.05 or P<0.01).Compared with the positive group,the serum AST level of rats in the middle dose group of Yiqi Yangyin Decoction was significantly lower(P<0.05),and the serum cGMP level of rats in the high dose group of Yiqi Yangyin Decoction was significantly lower(P<0.01).Conclusion The Self-proposed Yiqi Yangyin Decoction significantly improved the symptoms of rats of Qi and Yin deficiency syndrome,which may be related to the regulation of immune function and improvement of the cyclic nucleotide system.
