摘要
唑吡坦是一种国家二类精神药物,具有潜在的滥用风险。为了预防可能出现的安全事件,对涉及该药物的样本进行快速准确的检测至关重要。唑吡坦-d_(7)作为唑吡坦残留检测所需的重要内标化合物,其在分析检测中的重要性不言而喻。基于此背景,本文报道了一种合成唑吡坦-d_(7)的方法。以甲苯-d_(8)为起始原料,经过傅-克酰基化、环化反应、缩合反应、酯水解、黄鸣龙还原和酰胺化反应合成了唑吡坦-d_(7),该化合物结构经^(1)H NMR、^(13)C NMR和IR得到了进一步的确证,经HPLC分析纯度达到99.49%。
Zolpidem is a Class II controlled substance in China with potential abuse risks.In order to prevent possible safety incidents,it is critical to perform rapid and accurate testing of samples containing this drug.Zolpidem-d_(7)is an essential internal standard compound for detecting zolpidem residues and is undeniably significant in analytical testing.In this context,this paper presents a method for synthesizing zolpidem-d_(7).With toluene-d_(8)as the starting material,zolpidem-d_(7)was synthesized via the reaction of Friedel-Crafts acylation,cyclization,condensation,ester hydrolysis,Huang-Minlon reduction,and amidation.The structure was confirmed by^(1)H NMR,^(13)C NMR,and IR,and its purity was determined to be 99.49%by HPLC analysis.
