摘要
建立了同时测定中药制剂——三七片中三七皂苷R1、人参皂苷Rg1、人参皂苷Re、人参皂苷Rb1和人参皂苷Rd等5种皂苷类成分含量的高效液相色谱-蒸发光散射测定方法。样品采用超声辅助乙醇提取,通过安捷伦ZORBAXSB-C18(150 mm×4.6 mm,5μm)色谱柱和乙腈-水流动相梯度洗脱分离,蒸发光散射检测器进行检测,以外标标准曲线法定量。在优化的检测条件下,5种皂苷类成分分离良好,在各自范围内线性相关系数均大于0.9956,平均回收率为99.55%~104.90%,相对标准偏差为1.56%~2.79%。本方法可用于三七片5种皂苷类成分的同时测定。
A high performance liquid chromatography(HPLC)coupled with evaporative light scattering detection(ELSD)method is established for the simultaneous determination of 5 kinds of saponins in Sanchi tablets which are notoginsenoside Rl,ginsenoside Rgl,ginsenoside Re,ginsenoside Rb1 and ginsenoside Rd.The samples are extracted with ultrasonic-assisted ethanol,then separated on an Agilent ZORBAX SB-C18(150 mmx4.6 mm,5μm)column with the acetonitrile-water as the mobile phase by gradient elution,and detected by ELSD,finally quantified by external standard calibration curve method.Under these optimization conditions,5 kinds of saponins are separated well.The linear correlation coefficients are greater than 0.9956 within their own ranges.The average recoveries are in the range of 99.55%~104.90%,and the relative standard deviations are within 1.56%~2.79%.This method can be used for the simultaneous determination of 5 kinds of saponins in Sanchi tablets.
