基于原子转移自由基聚合反应的高灵敏度蛋白质印迹方法研究

摘要 目的探索基于金纳米颗粒(AuNPs)和原子转移自由基聚合反应(ATRP)的蛋白质印迹高灵敏度检测方法。方法将AuNPs通过ATRP附着到聚偏二氟乙烯(PVDF)膜上,通过PVDF膜对小分子的吸附作用,提高检测灵敏度。结果改进后的PVDF膜(PVDF-ATRP-AuNPs)对目标小分子的截留效率提高了80%左右,检测限显著降低。结论改进后的方法具有良好的稳定性、优异的灵敏度,能对超低浓度小分子待测物进行有效检测。 Objective To explore a highly sensitive Western blotting method based on gold nanoparticles(AuNPs)and atom transfer radical polymerization(ATRP).Methods AuNPs was attached to polyvinylidene fluoride(PVDF)membranes through ATRP,and the detection sensitivity was improved by the adsorption of small molecules by PVDF membrane.Results The improved PVDF membrane(PVDF-ATRP-AuNPs)increased the interception efficiency of target small molecules by about 80%and significantly reduced the limit of detection.Conclusion The improved method has good stability and excellent sensitivity,and can effectively detect ultra-low concentration small molecule analytes.
