摘要
为控制茯苓多糖散在提取过程中产生的主要杂质成分茯苓三萜的含量限度,采用香草醛-高氯酸比色法测定茯苓三萜物质总量。分别从测定波长、5%香草醛-冰醋酸溶液用量进行试验条件优化筛选;并进一步开展线性关系、精密度、稳定性、重复性、准确度方法学验证。优化结果显示,茯苓多糖测定最优试验条件为:波长为543 nm、5%香草醛-冰醋酸用量为0.4 mL。以吸光度为纵坐标(y),齐墩果酸含量(X)为横坐标作线性回归方程,得回归方程为:y=0.0064x-0.0462,R^(2)=0.9998(n=6),验证结果表明,齐墩果酸在36.26~217.56μg/mL范围内,吸光度与齐墩果酸含量呈良好的线性关系,平均回收率为97.60%。该检测方法灵敏度高,精密度、稳定性、重复性良好,且准确可靠,可作为茯苓三萜含量限度的控制方法。
In order to control the content limit of triterpenoids of Pachymaria polysaccharide powder produced in the extraction process,the total amount of triterpene was determined by vanillin perchloric acid colorimetric method.The experimental conditions were optimized from the wavelength and the dosage of 5%vanillin-glacial acetic acid solution.The method of linear relationship,precision,stability,repeatability and accuracy was further verified.The results showed that the optimal test conditions for the determination of poria polysaccharides were as follows:the wavelength was 543 nm and the dosage of 5%vanillin and glacial acetic acid was 0.4 mL.Taking absorbance as the vertical coordinate(y)and oleanolic acid content(X)as the horizontal coordinate for linear regression,the regression equation is as follows:y=0.0064x-0.0462,R^(2)=0.9998(n=6).The results showed that there was a good linear relationship between oleanolic acid absorbance and oleanolic acid content in the range of 36.26-217.56μg/mL,and the average recovery rate was 97.60%.The method has high sensitivity,precision,stability,repeatability,accuracy and reliability,and can be used as a control method for the content limit of triterpenes in Poria cocos.
