摘要
旨在探究辣蓼黄酮正丁醇部位(FNB)对小鼠氧化应激反应的调节作用。将72只SPF级小鼠随机分为空白对照组、伪狂犬病病毒(PRV)模型组、芦丁对照组,3个FNB处理组(25、50、100 mg/kg体重)。采用注射联合滴鼻的方式感染10^(2.33)TCID_(50)PRV,以建立小鼠体内氧化应激模型,病毒感染后1 h,用不同剂量的FNB灌胃给药,0.3 mL/只。连续灌胃7 d后采样测定氧化应激相关因子水平并进行病理组织学观察,期间记录小鼠的临床表现。结果显示,PRV感染后,小鼠脾脏、肺脏和肝脏发生了不同程度的病理损伤,FNB显著或极显著降低小鼠脾脏、心脏指数(P<0.05,P<0.01);FNB能增强血清中CAT、SOD、HO-1和NQO1的活性,升高血清中GSH、NO、T-AOC和脾脏中T-AOC的水平,剂量为100 mg/kg体重的FNB效果最为显著。结果表明,FNB具有良好抗氧化应激作用,并可有效缓解小鼠PRV感染引起的氧化应激对机体损伤。
The study was carried out to investigate the regulatory effect of N-butanol fraction of Polygonum hydropiper flavonoids(FNB)on oxidative stress in mice.Seventy-two SPF mice were randomly divided into blank control group,PRV model group,rutin control group,and three doses of FNB groups(25,50,100 mg/kg).The animals were infected with 10^(2.33) TCID_(50) PRV by injection combined with nasal drops to establish an oxidative stress model in mice,and 1 h later,three doses of FNB were administered by gavage,0.3 mL/mouse.After seven continuous days gavage,the levels of oxidative stress-related factors were measured and histopathological observation was performed,and the incidence of mice was recorded.The results showed that after PRV infection,the spleen,lung and liver were damaged to varying degrees,and FNB significantly or extremely significantly reduced indexes of spleen and heart of mice(P<0.05,P<0.01).The activities of CAT,SOD,HO-1 and NQO1 and the levels of GSH,NO and T-AOC in serum were increased and T-AOC in spleen was also raised.FNB treatment at 100 mg/kg revealed the best effect.The results showed that FNB has a good effect against oxidative stress and effectively alleviated the oxidative stress caused by PRV infection.
