膀胱癌组织及尿液外泌体中NTN1的表达与膀胱癌恶性程度的相关性研究

The correlation study of NTN1 expression in bladder cancer tissues and urine exosome with the progression of bladder cancer

摘要 目的探究NTN1在尿液和尿液外泌体中的表达对辅助诊断膀胱癌的潜在意义。方法应用TCGA-BLCA和GSE188715两个数据库中膀胱癌与癌旁差异基因,筛选出与差异性相关程度高的基因,构建膀胱癌的风险评分模型;在TCGA数据库中分析NTN1的表达与膀胱癌分级、分期、患者的生存期之间的相关性;ELISA的方法检测膀胱癌患者和非膀胱癌患者尿液及尿液外泌体中NTN1的含量。结果检索TCGA-BLCA和GSE188715两个数据库中膀胱癌与癌旁差异基因,筛选出膀胱癌发生的潜在重要风险因素NTN1和ACSM5,用这两个基因构建膀胱癌风险评分模型;在TCGA-BLCA数据库中膀胱癌的分期与分级越高,NTN1的表达越高,且NTN1在膀胱癌中的高表达与患者低生存率更相关。与非膀胱癌患者相比较,膀胱癌患者尿液及尿液外泌体中NTN1的含量更高,且膀胱癌分级分期越高的患者尿液外泌体中NTN1的含量越高。结论NTN1在膀胱癌组织中高表达,表达水平与膀胱癌分期与分级相关,NTN1的高表达预示着患者的不良预后;在膀胱癌患者尿液外泌体中NTN1的表达比在尿液中的表达更适合作为辅助膀胱癌诊断的潜在预测指标。 Objective To investigate the potential significance of NTN1 expression in urine and urinary exosomes for assisting in the diagnosis of bladder cancer.Methods The bladder cancer-related genes with the highest correlation were identified from the differentially expressed genes between bladder cancer and adjacent tissues in two databases,TCGA-BLCA and GSE188715,to construct a risk scoring model for bladder cancer using these genes.The TCGA database was used to analyze the correlation between NTN1 expression and bladder cancer staging,grading,and patient survival rates.Furthermore,the contents of NTN1 in urine and urinary exosomes of bladder cancer and non-bladder cancer patients were detected by ELISA.Results The results showed that NTN1 and ACSM5 were identified as potential important risk factors for bladder cancer occurrence based on the differentially expressed genes between bladder cancer and adjacent tissues in the TCGA-BLCA and GSE188715 databases,and a bladder cancer risk scoring model was constructed using these two genes.Moreover,the expression of NTN1 was positively correlated with bladder cancer staging and grading in the TCGA-BLCA database,and high expression of NTN1 was more strongly associated with low patient survival rates.In addition,the content of NTN1 in urine and urinary exosomes of bladder cancer patients was higher compared to non-bladder cancer patients,and the content of NTN1 in urinary exosomes had the potential to distinguish high and low stages of tumors.Conclusion NTN1 expression in urinary exosomes may be a more promising potential predictive indicator for assisting in the diagnosis of bladder cancer than its expression in urine.These findings may contribute to the development of a more efficient and accurate diagnostic approach for bladder cancer.
