摘要
目的探讨五味子乙素对骨肉瘤细胞增殖和凋亡的影响及潜在机制。方法体外培养骨肉瘤细胞系MG-63,给予五味子乙素(5、10、20、40和80μmol/L)处理,CCK-8实验和平板克隆实验检测细胞增殖能力,Annexin-V/PI染色实验检测细胞凋亡情况,Western blot检测细胞Bax、Bcl-2、Cleaved caspase 3、信号转导和转录激活因子3(STAT3)、p-STAT3蛋白表达。结果与对照组比较,不同浓度五味子乙素能显著降低MG-63细胞存活率,抑制细胞克隆形成,诱导细胞凋亡,促进细胞Bax与Cleaved Caspase-3蛋白表达,抑制Bcl-2蛋白表达(P<0.01);下调MG-63细胞p-STAT3蛋白表达(P<0.01)。结论五味子乙素可显著抑制骨肉瘤细胞MG-63增殖,诱导其凋亡,其机制可能与抑制STAT3信号通路激活有关。
Objective To investigate the effect of Schisandrin B on the proliferation and apoptosis of osteosarcoma cells and its potential mechanism.Methods Osteosarcoma cell line MG-63 was cultured in vitro and treated with Schisandrin B(5,10,20,40 and 80μmol/L mg/mL).CCK-8 assay and plate cloning assay were used to detect cell proliferation,and Annexin-V/PI staining was used to detect cell apoptosis.The expressions of Bax,Bcl-2,Cleaved caspase 3,signal transducer and activator of transcription 3(STAT3),and p-STAT3 were detected by Western blot.Results Compared with the control group,different concentrations of Schisandrin B significantly decreased the survival rate of MG-63 cells,inhibited cell cloning,induced cell apoptosis,upregulated the expression of Bax and Cleaved Caspase-3,inhibited the expression of Bcl-2(P<0.01).Schisandrin B significantly down-regulated the expression of p-STAT3 in MG-63 cells(P<0.01).Conclusion Schisandrin B could significantly inhibit the proliferation of osteosarcoma cells MG-63 and induce its apoptosis,which may be related to inhibiting the activation of STAT3 signaling pathway.
