毛报春叶片组培快繁体系建立

Establishment of rapid multiplication system for tissue culture of Primula×pubescens leaves

摘要 为了建立成熟的毛报春组织培养体系,以毛报春(Primula×pubescens)叶片为外植体,采用组培快繁的方法,研究了不同培养基、不同植物生长调节剂种类及浓度配比对愈伤组织诱导和分化、不定芽增殖及生根培养的影响,并对组织培养过程中出现的玻璃化苗现象进行恢复。结果表明:(1)最佳愈伤组织诱导培养基为MS+6.0 g/L琼脂+30.0 g/L蔗糖+1.2 mg/L NAA+1.5 mg/L 6-BA,诱导率可达96.67%,愈伤组织呈绿色,质地紧密。(2)愈伤组织分化最适培养基为MS+6.0 g/L琼脂+30.0 g/L蔗糖+0.5 mg/L NAA+2.0 mg/L 6-BA,分化出的丛生芽生长健壮。(3)最适不定芽增殖培养基为MS+6.0 g/L琼脂+30.0 g/L蔗糖+0.5 mg/L NAA+0.2 mg/L 6-BA,增殖率可达70.33%,苗绿且生长旺盛。(4)最有利于生根诱导的培养基为MS+6.0 g/L琼脂+30.0 g/L蔗糖+0.2 mg/L NAA,生根率可达70.67%,且根粗壮,不易脱落;(5)培养基中添加0.5 g/L活性炭或者3.0 mg/L AgNO 3玻璃化苗恢复效果最好。 In order to establish a mature Primula×pubescens tissue culture system,the method of tissue culture and rapid propagation was adopted,the effects of different culture media,different plant growth regulator types and concentration ratios on callus induction and differentiation,adventitious bud proliferation and rooting culture were studied by using Primula×pubescens leaves as explants,and the vitrification phenomenon in the tissue culture process was restored.The results showed that:(1)The optimal callus induction medium was MS+6.0 g/L agar+30.0 g/L sucrose+1.2 mg/L NAA+1.5 mg/L 6-BA,the induction rate could reach 96.67%,and the callus was green and compact in texture.(2)The most suitable medium for callus differentiation was MS+6.0 g/L agar+30.0 g/L sucrose+0.5 mg/L NAA+2.0 mg/L 6-BA,and the differentiated clumps of buds grew robustly.(3)The optimal adventitious bud proliferation medium was MS+6.0 g/L agar+30.0 g/L sucrose+0.5 mg/L NAA+0.2 mg/L 6-BA,the proliferation rate could reach 70.33%,and the seedlings were green and vigorously grown.(4)The most conducive medium for rooting-induced was MS+6.0 g/L agar+30.0 g/L sucrose+0.2 mg/L NAA,with a rooting rate of 70.67%,and the roots were thick and not easy to fall off.(5)The recovery effect of 0.5 g/L activated carbon or 3.0 mg/L AgNO 3 vitrified seedlings was the best in medium.
