摘要
随着高通量筛选和组合化学的不断发展,越来越多的新化学实体得以被发现,但这些新化学实体的研发进程常因其较差的水溶性而受限,这直接影响了药物在体内的生物利用度。近年来,为了提高难溶性药物的生物利用度,采用湿法研磨技术制备出更小粒径的纳米颗粒,已经成为研究的热点。本文综述湿法研磨效率的主要因素,纳米悬浮液的固化处理以及湿法研磨纳米制剂的质量评价,旨在为湿法研磨技术在纳米制剂中的应用提供理论基础和参考。
With the continuous development of high-throughput screening and combinatorial chemistry,more and more new chemical entities have been discovered,but the development process of these new chemical entities is often limited by their poor aqueous solubility,which directly affects the in vivo bioavailability of drugs.In order to improve the bioavailability of poorly soluble drugs,the preparation of nanoparticles with smaller particle size by wet milling has become a hot research topic in recent years.This paper reviews the efficiency of wet milling technology,solidification of nanoformulation and quality evaluation of nanoformualtion,aiming to provide a theoretical basis and reference for the application of wet milling technology in nanoformulation.
