摘要
目的 采用两样本孟德尔随机化方法研究精神分裂症、双相情感障碍、重性抑郁障碍、孤独症谱系障碍、注意缺陷与多动障碍的发病风险与大脑皮质厚度和表面积变化之间的因果关系。方法 在精神病基因组学联盟(Psychiatric Genomics Consortium)以及UK biobank分别获取精神分裂症、双相情感障碍、重性抑郁障碍、孤独症谱系障碍、注意缺陷与多动障碍大脑的皮质厚度和表面积全基因组关联研究(Genome wide association study,GWAS)分析结果,利用逆方差加权法(inverse variance weighted,IVW)产生主要结果,并采用MR-Egger和简单模型分析方法计算补充结果,此外还进行了敏感性分析。结果 发现以左侧颞横回表面积作为暴露因素、精神分裂症作为结局变量之间存在显著的因果关系(βIVW=-0.158、P<0.001)。结论 左侧颞横回表面积与精神分裂症存在单向因果关系(左侧颞横回表面积作为暴露因素)。
Objectives Two-sample Mendelian randomization was employed to investigate the causal relationship between alterations in cortical thickness and surface area and the risk of schizophrenia,bipolar disorder,major depressive disorder,autism spectrum disorder,attention deficit and hyperactivity disorder.Methods The results of Genome-wide Association Studies(GWASs)for schizophrenia,bipolar disorder,major depressive disorder,autism spectrum disorder,attention deficit and hyperactivity disorder,cortical thickness and surface area were downloaded from Psychiatric Genomics Consortium and the UK biobank.Inverse Variance Weighted(IVW)were used as the main results,and supplementary results were calculated using MR-Egger and Simple median analysis methods.In addition,sensitivity analysis was also performed.Results A significant causal relationship was found between the surface area of the left temporal transverse gyrus as exposure and schizophrenia as outcome.(βIVW=-0.158,P< 0.001).Conclusions A unidirectional causal relationship was identified between left transverse temporal gyrus surface area and schizophrenia(left transverse temporal gyrus surface area as exposure).
