lncRNA ITGA9-AS1、miR-670-3p在肝细胞癌组织中表达及与患者预后的关系

Expression of lncRNA ITGA9-AS1 and miR-670-3p in hepatocellular carcinoma tissues and their relationship with prognosis of patients

目的探究长链非编码RNA(lncRNA ITGA9-AS1)和miR-670-3p在肝细胞癌(HCC)组织中的表达及预后评估价值。方法以2016年6月至2018年6月邯郸市中心医院收治的76例肝细胞癌患者为研究对象,取其癌组织和相应癌旁组织,分别为HCC组和癌旁组。lncRNA ITGA9-AS1和miR-670-3p水平检测采用实时荧光定量PCR(qRT-PCR);Pearson相关性分析lncRNA ITGA9-AS1和miR-670-3p的关系;Kaplan-Meier法分析生存曲线;COX回归分析影响因素。结果HCC组lncRNA ITGA9-AS1水平低表达,miR-670-3p水平高表达(P<0.05)。lncRAN ITGA9-AS1与miR-670-3p呈负相关关系(r=-0.310,P=0.006)。lncRNA ITGA9-AS1、miR-670-3p表达水平与TNM分期、淋巴结转移、肿瘤分化、肿瘤大小、肿瘤数目有关(P<0.05)。随访3年发现lncRNA ITGA9-AS1高表达组患者累积生存率高于低表达组;miR-670-3p高表达组患者累积生存率低于低表达组(P<0.05)。多因素COX回归分析表明肿瘤分化(HR=1.842)、淋巴结转移(HR=3.882)、miR-670-3p(HR=3.786)、lncRNA ITGA9-AS1(HR=3.366)是影响HCC患者预后的因素(P<0.05)。结论HCC组织中lncRNA ITGA9-AS1呈低表达,miR-670-3p呈高表达,二者表达水平与临床病理特征存在密切关系,且对HCC患者预后有一定的评估价值。

Objective To explore the expression and prognosis evaluation value of long non-coding RNA(lncRNA ITGA9-AS1)and miR-670-3p in hepatocellular carcinoma(HCC)tissue.Methods Seventy-six patients with HCC who were admitted to our hospital from June 2016 to June 2018 were selected as the research objects,and their cancer tissues and corresponding paracancerous tissues were taken as the HCC group and the precancerous group,respectively.The levels of lncRNA ITGA9-AS1 and miR-670-3p were detected by real-time quantitative PCR(qRT-PCR);Pearson correlation was used to analyze the relationship between lncRNA ITGA9-AS1 and miR-670-3p;Kaplan-Meier method for analyzing survival curves;COX regression analysis of influencing factors.Results In HCC group,lncRNA ITGA9-AS1 level was low,and miR-670-3p level was high(P<0.05).lncRAN ITGA9-AS1 was negatively correlated with miR-670-3p(r=-0.310,P=0.006).The expression levels of lncRNA ITGA9-AS1 and miR-670-3p were correlated with TNM stage,lymph node metastasis,tumor differentiation,tumor size,and tumor number(P<0.05).After 3 years of follow-up,the cumulative survival rate of lncRNA ITGA9-AS1 patients in the high expression group was higher than that in the low expression group.The cumulative survival rate of patients with high expression of miR-670-3p was lower than that of patients with low expression(P<0.05).Multivariate COX regression analysis showed that tumor differentiation(HR=1.842),lymph node metastasis(HR=3.882),miR-670-3p(HR=3.786)and lncRNA ITGA9-AS1(HR=3.366)were the factors affecting the prognosis of HCC patients(P<0.05).Conclusion The lncRNA ITGA9-AS1 is lowly expressed and miR-670-3p is highly expressed in HCC tissue.The levels of the two are closely related to clinicopathological characteristics and have a certain value in evaluating the prognosis of HCC patients.
