摘要
食管癌是全球范围内常见的恶性肿瘤之一,具有较高的发病率和死亡率,尽管近年来在诊断和治疗技术上取得了显著进展,但食管癌的预后仍然较差,仍需揭示其机制和治疗靶点.BAG6(BCL2-associated Athanogene6)是一种多功能蛋白质,影响多种肿瘤发生发展,如结直肠癌、胃癌、肺癌等,然而,BAG6在食管癌中的具体作用和机制仍未被充分探索.本研究旨在探讨BAG6在食管癌中的表达及其生物学功能.通过分析食管癌组织和细胞系中BAG6的表达水平,并研究其过表达或敲低对食管癌细胞行为的影响,结果表明BAG6在食管癌组织中低表达.BAG6过表达会显著抑制食管癌细胞的生长、迁移,KYSE150和KYSE180细胞的增殖抑制率分别达到62%和58%,迁移抑制率分别为70%和65%.另外,BAG6过表达可抑制Wnt/β-catenin信号通路.这些发现表明BAG6作为潜在的肿瘤抑制因子,可能成为食管癌治疗的新靶点.
Esophageal cancer is one of the most common malignancies worldwide,with a high morbidity and mortality rate.Despite significant advances in diagnosis and treatment techniques in recent years,the prognosis of esophageal cancer is still poor,and its mechanisms and therapeutic targets still need to be revealed.BAG6(BCL2-Associated Athanogene 6)is a multifunctional protein,which affects the development of various tumors,such as colon cancer,gastric cancer and lung cancer.However,the specific role and mechanism of BAG6 in esophageal cancer have not been fully explored.The objective of this study was to investigate the expression and biological function of BAG6 in esophageal carcinoma.By analyzing the expression level of BAG6 in esophageal cancer tissues and cell lines,and studying the effect of overexpression or knockdown of BAG6 on the behavior of esophageal cancer cells,the results showed that BAG6was underexpressed in esophageal cancer tissues.Overexpression of BAG6 resulted in proliferation inhibition rates of 62%and 58%and migration inhibition rates of 70%and 65%in KYSE150 and KYSE180 cells,respectively,and inhibited the Wnt/β-catenin signaling pathway.These findings suggest that BAG6,as a potential tumor suppressor,may be a new target for the treatment of esophageal cancer.
