柞蚕高密度遗传图谱构建及体色性状QTL初步解析

摘要 为构建柞蚕高密度遗传图谱并对体色性状进行QTL初步解析,采用Super-GBS技术对115个柞蚕样品(2个亲本、3个F_(1)代个体、110个BC_(1)M个体)开展测序及基因分型,并利用区间作图法对体色性状进行QTL定位分析及候选基因发掘。结果得到柞蚕首张遗传图谱,全长4059.163 cM,平均遗传距离为1.17 cM,共有3466个标记分配到48个连锁群上;QTL定位分析共得到11个与体色性状相关的位点,分布在4个连锁群上,单个LOD范围1.92~16.86,其中在连锁群LG8与LG41上共获得9个QTL;在候选区间内进行基因挖掘,共得到11个候选基因。得到的候选基因为柞蚕体色性状调控机制研究提供了线索,助力柞蚕生物育种研究的开展。 To construct a high-density genetic map of Antheraea pernyi and preliminarily analyze QTLs for body color traits,115 tussah silkworm samples(2 parents,3 F 1 generation,and 110 BC 1 M)were sequenced and genotyped by Super-GBS technology.Interval mapping was used to QTL loci and to predict candidate genes.The results showed that the length of the first genetic map of Antheraea pernyi was 4059.163 cM,with an average genetic distance of 1.17 cM.A total of 3466 markers were assigned to 48 linkage groups,and 11 loci related to body color traits were obtained by QTL analysis,distributed in 4 linkage groups with a single LOD range of 1.92-16.86.A total of 9 QTLs were obtained on linkage groups LG8 and LG41,and 11 candidate genes were obtained by gene prediction and annotation in the candidate interval.The candidate genes obtained in this study provide insight into the regulatory mechanism of body color traits of Antheraea pernyi,contributing to the development of biological breeding research.
