摘要
以北沙参种子为试验材料,采用系统溶剂法对种子内源抑制物质进行分离和生物活性测定,同时结合气相色谱-质谱(GC-MS)联用仪分析其内源抑制物质的成分和含量。结果表明,北沙参初休及解除种子各有机相提取液对白菜种子的发芽率均存在抑制作用,且随着浸提液浓度的增加,抑制效果也不断增强;北沙参种子的各分离相提取液对白菜种子发芽能力的作用依次为:石油醚相>水相>二氯甲烷相>乙酸乙酯相>甲醇相;北沙参种子的各分离相提取液对白菜种子的抑制作用依次为:甲醇相>二氯甲烷相>乙酸乙酯相>水相>石油醚相;北沙参种子休眠的抑制作用大于非休眠。对各分离相分离鉴定发现,北沙参种子中含有种类较多的有机化合物,多为有机酸类、香豆素类、酯类、酚类、醛类、萜类、醇类和烃类物质等。北沙参种子中存在的内源抑制物是导致其休眠的主要原因之一。
In this paper,using Glehnia littoralis seeds as research materials,of which the seed endogenous inhibitory substances were isolated and their bioactivity was determined by systematic solvent method,and the composition and content of the endogenous inhibitory substances were analyzed by gas chromatography-mass spectrometry(GC-MS).The results showed that the germination rate of Chinese cabbage seeds was inhibited by the organic phase extracts from the early resting and the discharged Glehnia littoralis seeds,and the inhibitory effect was enhanced with the increase of the concentration of the extracts.The effects of each phase extract on the germination ability of Chinese cabbage seeds were as follows:petroleum ether phase>water phase>dichloromethane phase>ethyl acetate phase>methanol phase.The inhibitory effects of the extracts of the seeds were as follows:methanol phase>dichloromethane phase>ethyl acetate phase>water phase>petroleum ether phase.The inhibitory effect of seed dormancy was greater than that of non-dormancy.It was found that the seeds contained many kinds of organic compounds,such as organic acids,coumarins,esters,phenols,aldehydes,terpenes,alcohols and hydrocarbons.The existence of endogenous inhibitors in the Glehnia littoralis seeds was one of the main causes of dormancy.
