甜菜小G蛋白BvRAC基因家族鉴定及生物信息学分析

摘要 小G蛋白RAC(RAS-related C3 botulinum toxin substrates)基因家族参与植物生长、发育及逆境胁迫调控过程,是植物信号转导领域的研究热点。为了探究甜菜BvRAC基因家族的功能与进化关系,利用生物信息学方法对BvRAC基因家族成员进行鉴定,并对其理化性质、多序列比对、系统进化、基因结构、染色体定位及共线性、启动子顺式作用元件、盐胁迫下的表达模式和蛋白互作网络进行研究。分析结果表明,BvRAC基因家族有9个成员,有6~8个外显子,具有5个G结构域(G1~G5),1个SYR基序(S为Ser,Y为Tyr,R为Arg)、1个Rho插入序列和1个C端可变区。BvRAC基因家族成员的启动子中含有多个激素和逆境应答元件;8个基因均响应盐胁迫;BvRAC蛋白通过与鸟苷酸交换因子(GDP exchange factor,GEF)、GTP酶激活蛋白(GTPase activating protein,GAP)、鸟苷酸解离抑制因子(GDP dissociation inhibitor,GDI)、含有CRIB结构域的ROP相互作用蛋白(ROP-interactive CRIB-containing Protein,RIC)、蛋白磷酸酶2C(Protein phosphatase 2C,PP2C)等蛋白互作发挥调节甜菜生长及逆境应答功能。本研究为甜菜小G蛋白BvRAC基因家族在甜菜生长发育和逆境胁迫下功能研究奠定了良好的基础。 The small GTP-binding proteins RAS-related C3 botulinum toxin substrates(RAC)gene family is involved in the regulation of plant growth,development and adversity stress,and is a hot research topic in the field of plant signal transduction.In order to investigate the functional and evolutionary relationships of the RAC gene family from sugar beet,the bioinformatics methods were used to identify the BvRAC gene family members and investigate their physicochemical properties,multiple sequence comparison,phylogeny,gene structure,chromosomal localisation and co-linkage,promoter cis-acting elements,expression patterns under salt stress,and protein-interaction networks.The results show that there are 9 members in the BvRAC gene family,which have 6~8 exons with G domains,SYR motifs(S is Ser,Y is Tyr,R is Arg),Rho insert region and C-terminal HVR;the promoters of the BvRAC gene family members contain several hormones and stress responsive elements;all eight genes respond to salt stress;BvRAC proteins regulate sugar beet growth and stress response structure by interacting with proteins such as GDP exchange factor(GEF),GTPase activating protein(GAP),GDP dissociation inhibitor(GDI),ROP-interactive CRIB-containing protein(RIC),protein phosphatase 2C(PP2C)and other proteins.This study has laid a solid foundation for the functional study of the BvRAC gene family of sugar beet small GTP-binding protein under growth,development,and stress conditions.
