霍山石斛多糖抑制帕金森病模型神经炎性损伤作用研究

Study on the inhibitory effect of Dendrobium Huoshan polysaccharide on neuroinflammatory damage in Parkinson's disease model

摘要 目的研究霍山石斛多糖(DHP)对帕金森病(PD)模型神经炎性损伤的作用。方法2022年6月至2023年2月,将小胶质细胞BV-2分为正常组(不做任何干预正常培养)、模型组[以1-甲基-4-苯基吡啶离子(MPP+)诱导]、DHP组(先行2~400 mg/L DHP干预,后加模型组相同剂量MPP+诱导),应用酶联免疫吸附测定(ELISA)检测细胞白细胞介素(IL)-1β含量;30只C57BL/6小鼠采用随机数字表法分为正常组、模型组、DHP组,每组10只。设置DHP等效治疗剂量即350 mg/kg为DHP组。造模完成后,正常组和模型组灌胃双蒸水0.5 mL,1次/天,持续2周;DHP组灌胃0.5 mL DHP溶液,浓度为15.4 g/mL,1次/天,持续2周。通过平衡木实验和转棒试验观察小鼠平衡能力、肌力以及运动协调力;ELISA检测小鼠中脑组织IL-1β和IL-18炎症因子表达水平;免疫荧光染色检测小鼠中脑黑质离子钙结合衔接分子1(Iba-1)和黑质酪氨酸羟化酶(TH)表达水平。结果实验结果显示,与正常组比较,BV-2细胞模型组IL-1β水平显著升高[(76.32±6.18)ng/L比(51.12±4.72)ng/L,P<0.05];与模型组比较,DHP组IL-1β水平降低(P<0.05),且在2.0~100.0 mg/L内呈剂量依赖性下降。与正常组比较,模型组通过平衡木时间显著延长(P<0.05),在棒时间缩短(P<0.05),中脑组织IL-1β[(190.10±14.68)ng/L比(80.46±15.18)ng/L]和IL-18水平升高(P<0.05);与模型组比较,DHP组通过平衡木运动时间缩短(P<0.05),在棒时间延长(P<0.05),DHP组中脑组织IL-1β[(89.69±14.31)ng/L]和IL-18水平降低(P<0.05)。免疫荧光结果显示,与正常组比较,模型组中脑黑质Iba-1表达显著升高(P<0.05),TH表达下降(P<0.05);与模型组比较,DHP组中脑黑质Iba-1表达显著降低(P<0.05),TH表达升高(P<0.05)。结论DHP可能通过抑制小胶质细胞过度活化,进而降低PD模型神经炎性损伤,发挥神经保护作用。 Objective To investigate the neuroprotective effects of polysaccharides from Dendrobium huoshanense(DHP)on neuroinflammatory damage in a Parkinson's disease(PD)model.Methods BV-2 microglial cells were divided into three groups:normal control(NC,no intervention for normal culture),model(Mod,Induced with MPP+),and DHP-treated(first 2 to 400 mg/L DHP intervention,plus the same dose of MPP+induction in the model group)group.The levels of interleukin-1β(IL-1β)in cells were measured using enzyme-linked immunosorbent assay(ELISA).Thirty C57BL/6 mice were randomly assigned to three groups(n=10 per group):NC,Mod,and DHP.The DHP group received an equivalent dose of 350 mg/kg DHP.After modeling,the NC and Mod groups received daily gavage with 0.5 mL ddH2O for two weeks,while the DHP group received 0.5 mL of a 15.4 g/L DHP solution daily for two weeks.Balance beam and rotarod tests were used to assess the mice's balance,muscle strength,and motor coordination.Levels of inflammatory cytokines IL-1βand IL-18 in midbrain tissue were measured by ELISA,while immunofluorescence staining was used to evaluate expression levels of ionized calcium-binding adapter molecule 1(Iba-1)and tyrosine hydroxylase(TH)in the substantia nigra.Results Com‑pared with the NC group,IL-1βlevels in the BV-2 cell model group were significantly increased[(76.32±6.18)ng/L vs.(51.12±4.72)ng/L,P<0.05].In comparison with the Mod group,IL-1βlevels in the DHP group decreased in a dose-dependent manner within the 2.0-100.0 mg/L range(P<0.05).Compared to the NC group,the Mod group showed a significant increase in balance beam time and a reduction in rotarod time(P<0.05),with elevated levels of IL-1β[(190.10±14.68)ng/L vs.(80.46±15.18)ng/L]and IL-18 in midbrain tissue(P<0.05).Compared to the Mod group,the DHP group demonstrated a reduction in balance beam time(P<0.05),an increase in rotarod time(P<0.05),and decreased IL-1β[(89.69±14.31)ng/L]and IL-18 levels(P<0.05).Immunofluorescence results showed that compared with the NC group,Iba-1 expression in the substantia nigra of the Mod group was significantly increased(P<0.05),while TH expression decreased(P<0.05).Compared to the Mod group,the DHP group showed a significant reduction in Iba-1 expression and an increase in TH expression(P<0.05).Conclusion DHP may exert neuroprotective effects by inhibiting excessive activation of microglia,thereby reducing neuroinflammatory damage in PD models.
