摘要
目的:研究槲皮素(quercetin,QUE)对胃癌的抑制及免疫调节作用。方法:通过CCK-8法(cell counting Kit-8,CCK-8)检测不同浓度QUE处理后HGC-27细胞存活率,筛选有效作用浓度;苏木精-伊红染色观察25μM组、50μM组处理24 h后HGC-27细胞形态;流式细胞术检测25μM组、50μM组处理24 h后细胞周期及凋亡变化;流式细胞术检测25μM组、50μM组处理后小鼠外周血中免疫细胞变化。结果:与对照组比较,25μM组、50μM组能够有效抑制HGC-27细胞增殖活性(P<0.01);50μM组HGC-27细胞质空泡化,G_(0)/G_(1)期缩短(P<0.001),S期延长(P<0.001),G_(2)/M期消失(P<0.001);50μM组中细胞凋亡率增加(P<0.001);50μM组小鼠外周血中T淋巴细胞比例增加(P<0.001)。结论:QUE可以在S期阻滞HGC-27细胞增殖,促进细胞凋亡;其中高浓度QUE对于胃癌瘤体的抑制作用显著,QUE能够通过增强机体免疫功能发挥抑制肿瘤作用。
Objective:To study the inhibitory effects of quercetin(QUE)on gastric cancer and its immuno-modulation mechanism.Methods:The survival rate of HGC-27 cells after treatment with different concentrations of QUE was detected by CCK-8 to screen the effective concentrations;the morphology of HGC-27 cells was observed by HE staining method after 24 h of treatment in 25μM and 50μM groups;flow cytometry was used to detect the cell cycles and apoptosis after 24 h of treatment in 25μM and 50μM groups;and the changes in immune cells in the peripheral blood of mice after 24 h treatment in the 25μM and 50μM groups.Results:Compared with the control group,25μM and 50μM groups could effectively inhibit the activity of HGC-27 cells(P<0.01);HGC-27 cytoplasm became vacuolated after 50μM QUE treatment,G_(0)/G_(1)phase shortened(P<0.001),the S phase prolonged(P<0.001),the G_(2)/M phase disappeared(P<0.001);cellular apoptosis increased remarkably in the 50μM group(P<0.001);the increase in the proportion of T cells in the peripheral blood of the mice after 50μM QUE treatment could be found(P<0.001).Conclusion:QUE could block the proliferation of HGC-27 cells in the S phase and promote cellular apoptosis;among them,the high concentration of QUE has the significant inhibitory effects on the tumor of gastric cancer,QUE could inhibit tumor by enhancing the immune function of the body.
