全蝎烫伤油对小鼠深Ⅱ度烫伤的治疗作用及机制

摘要 目的 探究全蝎烫伤油对小鼠深Ⅱ度烫伤的治疗作用及其促进创面愈合的作用机制。方法 健康6~8周龄SPF级Balb-c小鼠140只,雌雄各半,体质量18~23 g,采用随机数字表法分为烫伤模型组、全蝎烫伤油组、磺胺嘧啶银组和湿润烧伤膏组,每组35只。建立小鼠深Ⅱ度烫伤模型,造模2 h清创后,烫伤模型组予0.9%氯化钠注射液涂抹,其他药物干预组予相应药物0.5 g均匀涂抹于创面,每日早晚各换药1次。测定创面愈合率、创面愈合时间,免疫组化法测定创面组织微血管密度(MVD)。结果 在给药后第7天、第14天、第21天全蝎烫伤油组创面愈合率高于同时间点烫伤模型组和其他2个药物干预组,差异具有统计学意义(P <0.05)。全蝎烫伤油组创面愈合时间明显短于烫伤模型组和其他2个药物干预组,差异具有统计学意义(P <0.05)。给药后第7天、第14天、第21天与同时间点烫伤模型组和其他2个药物干预组比较,全蝎烫伤油组创面组织MVD显著增多,差异具有统计学意义(P <0.05)。结论 全蝎烫伤油对小鼠深Ⅱ度烫伤模型具有良好的保护作用,可能与促进创面微血管形成,从而有效加速创面的愈合和修复有关。 Objective To explore the therapeutic effect of scorpion scald oil on deep second-degree scald in mice and its mechanism of promoting wound healing.Methods A total of 140 healthy SPF-grade Balb-c mice,aged 6-8 weeks,with a body weight of 18-23 g,were randomly divided into the scald model group,scorpion scald oil group,sulfadiazine silver group,and MEBO group,with 35 mice in each group.The model of deep second-degree scald in mice was established.After 2 hours of debridement,the scald model group was treated with 0.9%saline,while the other drug intervention groups were treated with 0.5 g of the corresponding drug,evenly applied to the wound surface.Dressings were changed once in the morning and evening.The wound healing rate and wound healing time were measured.The microvessel density(MVD)of scalded wound tissue was measured by immunohistochemical method.Results On the 7th,14th,and 21st days after administration,the wound healing rate in the scorpion scald oil group was significantly higher than that in the scald model group at the same time points(P<0.05).The wound healing time in the scorpion scald oil group was significantly shorter than that in the scald model group(P<0.05).On the 7th,14th,and 21st days after administration,the MVD in the scorpion scald oil group was significantly higher than that in the scald model group(P<0.05).Conclusion Scorpion scald oil has a good protective effect on deep second-degree scald in mice,which may be related to its promotion of wound microangiogenesis,effectively accelerating wound healing and repair.
