摘要
以茵陈蒿水提物(Aqueous Extract of Artemisia capillaris Thunb.,AEACT)为研究对象,探讨其对副溶血性弧菌(Vibrio parahaemolyticus,VP)的抑菌效果及其作用机制。采用微量稀释法测定VP的最小抑菌浓度(Minimal Inhibitory Concentration,MIC);以比浊法测定AEACT处理后VP的生长曲线;使用酶标仪测定电导率值;使用扫描电镜观察细菌外观形态变化。研究结果显示,AEACT能显著抑制VP的生长,其MIC为0.10 mg·mL^(-1);AEACT对VP生物膜的形成具有抑制和清除作用;AEACT处理后的细菌悬浮液电导率值有显著提升;扫描电镜观察显示,VP表面出现破裂和皱缩等损伤形态。综上所述,AEACT对VP具有显著的抑制作用,其机制可能与影响细胞膜的通透性有关。
Aqueous extract of Artemisia capillaris Thunb.(AEACT) was used as the research object to explore its antibacterial effect and mechanism of action against Vibrio parahaemolyticus(VP).The minimum inhibitory concentration(MIC) of VP were determined using the microdilution method;the growth curve of VP after AEACT treatment was determined using the turbidimetric method;the conductivity was determined using a microplate reader;use scanning electron microscopy to observe changes in bacterial appearance.The results show that AEACT can significantly inhibit the growth of VP,with an MIC of 0.10 mg·mL^(-1);AEACT has an inhibitory and clearing effect on the formation of VP biofilm;bacteria after AEACT treatment the conductivity value of the suspension was significantly improved;scanning electron microscope observation showed that the surface of VP showed damage patterns such as rupture and shrinkage.In summary,AEACT has a significant inhibitory effect on VP,and its mechanism may be related to affecting the permeability of the cell membrane.
