摘要
目的:观察小儿豉翘清热颗粒联合阿奇霉素对肺炎支原体感染大鼠的干预效果。方法:60只SPF级大鼠随机分为正常组、模型组、阿奇霉素组、小儿豉翘清热颗粒组、中西药结合组(小儿豉翘清热颗粒+阿奇霉素),每组12只。除正常组外,其余各组大鼠构建支原体肺炎模型。造模后,各组连续给药5 d,给药结束次日对各组大鼠进行腹主动脉取血,ELISA法检测血清IL-6、TNF-α及补体C3浓度,并取肺组织进行HE染色观察。结果:造模后,除正常组,其余各组大鼠体重均下降,且与正常组体重差值比较,差异均有统计学意义;治疗后,模型组体重进一步下降,其余各药物组治疗后体重均上升,且体重差值与模型组比较差异有统计学意义,中西药结合组治疗前后体重差值最大(P<0.05)。与正常组比较,其余各组大鼠血清中IL-6、TNF-α及补体C3浓度均升高;与模型组比较,各给药组大鼠血清中IL-6、TNF-α及补体C3浓度均下降,且中西药结合组下降趋势更明显(P<0.05)。病理学结果显示阿奇霉素和小儿豉翘清热颗粒均能部分缓解肺部炎症症状,而中西药结合的缓解作用更为明显。结论:小儿豉翘清热颗粒联合阿奇霉素可降低肺炎支原体感染大鼠血清IL-6、TNF-α及补体C3浓度,缓解肺部炎症症状。
Aim:To observe the intervention effects of Xiaoer Chiqiao Qingre granule combined with azithromycin on rats with Mycoplasma pneumoniae infection.Methods:Sixty SPF rats were randomly allocated into normal group,model group,Xiaoer Chiqiao Qingre granule group,azithromycin group and combination group(Xiaoer Chiqiao Qingre granule+azithromycin),with 12 rats in each group.Except the normal group,Mycoplasma pneumoniae pneumonia model was established in other 4 groups.Each group was administered continuously for 5 days.On the next day after the end of administration,blood was collected from the abdominal aorta of rats in each group.The serum concentrations of IL-6,TNF-αand complement C3 were determined by ELISA.Moreover,lung tissue was collected for HE staining.Results:After modeling,except the normal group,the body weight of the rats in the other 3 medication groups decreased,and the difference was statistically significant compared with the normal group(P<0.05).After treatment,the body weight of the model group decreased further,and the body weight of the other medication 3 groups increased after treatment,and the difference was statistically significant compared with the model group(P<0.05).The difference of body weight before and after treatment was the largest in the combination group(P<0.05).The serum levels of IL-6,TNF-αand complement C3 in the other 4 groups were significantly increased compared with the normal group(P<0.05).Compared with the model group,the serum levels of IL-6,TNF-αand complement C3 in the 3 medication groups especially the combination group were significantly decreased(P<0.05).Moreover,the results of pathological test suggested that both azithromycin and Xiaoer Chiqiao Qingre granule could partially relieve the symptoms of pulmonary inflammation,and the combination of the 2 drugs had more pronounced effect.Conclusion:Combination of Xiaoer Chiqiao Qingre granule and azithromycin could reduce serum levels of IL-6,TNF-αand complement C3,and relieve the symptoms of lung inflammation in rats with Mycoplasma pneumoniae infection.
