摘要
现有商用聚合酶链反应(PolymeraseChainReaction,PCR)仪以金属孔板作为控制对象,为了使管内样本温度维持足够时间,常以延长程序设定时间来对冲管内温度滞后对检测结果的影响,但会导致实验耗时长、管内温度不精确等后果.本文提出一种利用导热金属孔板过冲热量抵消PCR管内温度滞后的方法对管内温度进行修正,设计开发热循环控制系统,并用非洲猪瘟模板进行实验验证.结果表明,PCR管内温度校正后升降温耗时分别缩短了56.7%和28.6%,PCR扩增曲线Ct值提前了2.64±0.48,有效提升了PCR的检测效率及结果可靠性.
The existing commercial polymerase chain reaction(PCR)instrument utilizes the metal hole plate as the control object.In order to maintain the sample temperature in the tube for a sufficient length of time,the program setting time is often extended to reduce the impact of the temperature lag in the tube on the detection results.The method has problems such as long experimental time and inaccurate temperature in the tube.This paper proposes a method to correct the temperature by offsetting the temperature lag in the PCR tube.The thermal cycle control system was designed,and the African swine fever(ASF)template was used for experimental verification.The results showed that after the temperature correction in PCR tube,the time of temperature rising and cooling was reduced by 56.7%and 28.6%respectively,and the Ct value of the PCR amplification curve was advanced by 2.64±0.48,effectively improving the detection efficiency and reliability of PCR.
