膀胱癌组织中miR-451表达及其对患者术后生存预后的预测价值分析

Expression of miR-451 in bladder cancer tissues and its predictive value for postoperative survival and prognosis

目的探讨膀胱癌组织中微小RNA-451(miR-451)的表达及其对患者术后生存预后预测价值。方法选择2019年1月至2021年1月本院行根治性切除术的95例膀胱癌患者为研究对象,收集患者的癌组织及癌旁组织。采用实时荧光定量聚合酶链式反应(qRT-PCR)测定组织中的miR-451相对表达量。所有膀胱癌患者均随访至2023年1月,记录患者的总生存期(OS)。比较癌组织与癌旁组织中的miR-451相对表达量以及不同病理特征患者癌组织中的miR-451相对表达量;分析不同miR-451表达水平的OS差异;采用多因素logistic回归分析影响术后生存的独立危险因素。结果癌组织中的miR-451相对表达量低于癌旁组织(P<0.05)。临床分期为Ⅲ期的miR-451相对表达量低于Ⅰ~Ⅱ期(P<0.05);淋巴结转移患者癌组织中的miR-451相对表达量低于无淋巴结转移(P<0.05)。miR-451高表达患者的OS高于miR-451低表达患者(P<0.05)。多因素logistic回归分析结果显示,Ⅲ期、淋巴结转移和miR-451低表达为影响膀胱癌术后生存的独立危险因素。结论miR-451低表达在膀胱癌患者的疾病发生、发展中发挥着重要的作用,且与患者术后生存预后相关,也是影响术后生存的独立危险因素。

ObjectiveTo investigate the expression of microrna-451(miR-451)in bladder cancer tissues and its predictive value for postoperative survival and prognosis.MethodsFrom January 2019 to January 2021,95 patients with bladder cancer who underwent radical resection in our hospital were selected,and the cancer group and adjacent tissues were collected.The relative expression of miR-451 was measured by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).All patients with bladder cancer were followed up to January 2023,and the total overall survival(OS)was recorded.The relative expression of miR-451 in cancer tissue and adjacent tissues were compared.The relative expressiones of miR-451 in different pathological characteristics were compared.The OS differences of miR-451 expression levels were analyzed.Multivariate logistic regression analysis was used to analyze independent risk factors for postoperative survival.ResultsThe relative expression of miR-451 in cancer tissues was lower than that in adjacent tissues(P<0.05).The relative expression level of miR-451 in stageⅢwas lower than that in stageⅠtoⅡ(P<0.05).The relative expression of miR-451 in lymph node metastasis was lower than that without lymph node metastasis(P<0.05).The OS with high expression of miR-451 was longer than that with low expression of miR-451(P<0.05).Multivariate logistic regression analysis showed that stageⅢ,lymph node metastasis and low expression of miR-451 were independent risk factors for postoperative survival.ConclusionsThe low expression of miR-451 in bladder cancer patients is closely related to the survival and prognosis of patients after surgery,and the low expression of miR-451 is an independent risk factor for survival after surgery.
