肾细胞癌组织中PRL-3、CNNM3蛋白表达水平与患者临床表现、随访预后的关系

Relationship between PRL-3 and CNNM3 protein expression levels in renal cell carcinoma tissues and clinical manifestations and follow-up prognosis of patients

摘要 目的分析肾细胞癌组织中肝细胞再生磷酸酶3(PRL-3)、周期素M3(CNNM3)蛋白表达水平与患者临床表现、随访预后的关系。方法收集2015年6月至2018年6月在本院确诊并进行手术治疗的102例肾细胞癌患者的癌组织及癌旁组织标本。采用免疫组化法检测组织中PRL-3、CNNM3的蛋白表达水平。分析PRL-3、CNNM3与肾细胞癌患者临床病理特征的关系。通过多因素Cox回归分析探讨影响肾细胞癌患者预后的相关因素。结果肾细胞癌组织中的PRL-3、CNNM3阳性表达率(63.73%、58.82%)明显高于癌旁组织(39.22%、34.31%)(均P<0.05)。不同临床分期、病理分级、淋巴结转移肾细胞癌患者组织中的PRL-3、CNNM3蛋白表达水平比较,差异均有统计学意义(均P<0.05)。PRL-3阳性患者、CNNM3阳性患者的5年生存率(43.08%、45.00%)分别低于PRL-3阴性、CNNM3阴性患者(91.89%、83.33%)(均P<0.05)。多因素Cox回归分析显示,PRL-3阳性(HR=3.575,95%CI:2.037~6.275)、CNNM3阳性(HR=4.250,95%CI:2.078~8.692)均是影响肾细胞癌患者预后的危险因素(均P<0.05)。结论肾细胞癌组织中PRL-3、CNNM3蛋白主要呈阳性表达,PRL-3、CNNM3阳性表达的肾细胞癌患者的淋巴结转移、肿瘤分期晚期、病理高分级风险更大,预后更差,二者有望成为评估肾细胞癌临床病理特征及预后的有效生物标志物。 ObjectiveTo explore the relationship between the expression levels of hepatocyte regenerative phosphatase 3(PRL-3)and cyclin M3(CNNM3)protein in renal cell carcinoma tissues and the clinical manifestations and follow-up prognosis of patients.MethodsThe tumor tissues and adjacent tissues of 102 patients with renal cell carcinoma diagnosed and operated in our hospital from June 2015 to June 2018 were collected.The protein expression levels of PRL-3 and CNNM3 were detected by immunohistochemical method.The relationship between the PRL-3,CNNM3 and the features of renal cell carcinoma were explored.Cox regression analysis were used to investigate the relationship between PRL-3 and CNNM3 protein expression and the prognosis of patients with renal cell carcinoma.ResultsThe positive expression rates of PRL-3 and CNNM3 in renal cell carcinoma tissues were significantly higher than those in para-carcinoma tissues(all P<0.05).The protein expression levels of PRL-3 and CNNM3 in renal cell carcinoma tissues were significantly correlated with clinical stage,pathological grade and lymph node metastasis(all P<0.05).The 5-year survival rate(43.08%)in patients with positive expression of PRL-3 was lower than that in cases with negative expression(91.89%),and the 5-year survival rate(45.00%)in cases with positive expression of CNNM3 was lower than that in cases with negative expression(83.33%)(all P<0.05).Positive expression of PRL-3(HR=3.575,95%CI:2.037-6.275)and positive expression of CNNM3(HR=4.250,95%CI:2.078-8.692)were risk factors for prognosis of renal cell carcinoma(all P<0.05).ConclusionsPRL-3 and CNNM3 are highly expressed in renal cell carcinoma tissues,and patients with positive expression of PRL-3 and CNNM3 have higher risk of lymph node metastasis,advanced tumor stage,and high pathological grade,and worse prognosis.PRL-3 and CNNM3 are expected to be effective biomarkers for evaluating clinicopathological features and prognosis of renal cell carcinoma.
