基于网络药理学研究益肾结透汤治疗慢性肾脏病的机制

目的运用网络药理学分析益肾结透汤延缓慢性肾脏病(CKD)进展的潜在作用机制。方法运用TCMSP数据库及BATMAN-TCM数据库,筛选出益肾结透汤具有潜在药效的活性成分,通过运用PubChem数据库及Swiss Target Prediction平台进行靶点的预测分析,筛选药物的潜在靶点,运用OMIM及GeneCard数据库获取CKD的疾病靶点,筛选药物及疾病的交集靶点并构建PPI网络图获取核心靶点,通过Cytoscape软件构建药物—活性成分—靶点等多层次网络关联图,通过degree值获取核心化学成分;利用DAVID数据库对所获取的交集靶点进行GO功能及KEGG通路富集分析并将结果可视化。结果共筛选到162个活性成分,交集靶点168个,主要作用在AKT1、Alb、TNF等核心靶点,预测其可能通过AGE-RAGE、MAPK等信号通路发挥作用。结论益肾结透汤治疗CKD是以其中药物的核心成分槲皮素、光甘草定等,通过AGE-RAGE、MAPK等信号通路作用于AKT1、Alb、TNF等核心靶点,表明益肾结透汤治疗CKD具有多途径、多靶点作用的特点,为益肾结透汤治疗CKD提供了理论依据,为进一步的临床试验提供了参考依据。

Objective The potential mechanism of Yishen Jietou decoction delaying the progression of chronic kidney disease(CKD)was analyzed by network pharmacology.Methods TCMSP database and BATMAN-TCM database were used to screen out the active ingredients of Yishen Jietou decoction with potential efficacy,and the Target Prediction analysis was carried out through PubChem database and Swiss Target Prediction platform.To screen potential targets of drugs,obtain disease targets of CKD using OMIM and GeneCard databases,screen intersection targets of drugs and diseases,and construct PPI network diagram to obtain core targets.Construct drug-active ingredient-target and other multi-level network correlation diagram via Cytoscape software,and obtain core chemical components by degree value.GO function and KEGG pathway enrichment analysis were performed on the intersection targets obtained by using DAVID database and the results were visualized.Results A total of 162 active ingredients,were screened,and 168 intersection targets were identified,which mainly acted on core targets such as AKT1,Alb,TNF,etc.It was predicted that they might play their roles through signaling pathways such as AGE-RAGE and MAPK.Conclusion The treatment of CKD by Yishen Jietou decoction is based on quercetin,glycyrrhizin and other core drug components,and acts on AKT1,Alb,TNF and other core targets through AGE-RAGE,MAPK and other signaling pathways,indicating that Yishen Jietou decoction has the characteristics of multi-pathway and multi-target action in the treatment of CKD,providing a theoretical basis for Yishen Jietou decoction to treat CKD.It provides a reference for further clinical trials.
